Purpose: To study the ameliorated effect of recombinant growth hormone(rhGH) on protein synthesis in patients with liver cirrhosis pre- and-postoperatively, and its mechanism on protein synthesis. Methods: Forty patients were divided randomly into groups (control group, preoperative rhGH group, postoperative rhGH group, pre-and- postoperative rhGH group). Total protein, albumin, prealbumin, transferrin, AST, ALT, electrolyte, inosine, urea nitrogrn and blood sugar were detected in the mornings of preoperative dayO, day8(operation day) and postoperative day 1, day8. The liver samples were obtained during operation and preserved in fluid nitrogen promptly. After abstracting the total RINA from samples respectively and transforming mRNA to cDNA by reverse transcript technique, the albumin mRNA was amplified in the PCR system and determined semi-quantitatively with agarose gel electrophoresis. Result: 1. The serum AST, ALT, electrolytes, inosine, urea nitrogen and blood sugar were no difference in the four groups. 2. The serum total protein, albumin, peralbuinin, transferrin in the rhGH groups increased highly than those in the control group on preoperative day8 and postoperative day8. 3. Albumin mRNA in the rhGH groups was higher those in the control group. 4. The patients in rhGH groups felt better than those in the control group. 5. Postoperative complications in the rhGH groups were less than those in the control group. 6. Hospitalizing duration in the rhGH groups was shorter than those in the control group. 7. There -3- were no obvious hyperglycemia and body fluid retention. Conclusions: Albumin niRNA content markedly increased by rhGH. The rhGH can ameliorate protein synthesis and hypoproteinemia, and was favorable for prognosis.
