| THE STUDY OF PATHOLOGY AND HSP70 EXPRESSION INTERFERED BY DRUGS OF HYPERTENSIVE RATS AFTER MCAO AND REPERFUSIONABSTRACTObjectives To study the pathology of acute cerebral ischemic reperfusion damage and the affection of some drug to the expression of HSP70 and its cerebral protection afterischemic reperfusion. Methods Rats were distributed to two groups: 1.Hypertension group: Double renal artery stenosis of the rats were performed. After this operation, the rats of this group were fed for two months. Then measured their blood pressure. The rats whose blood pressure were high were separated into three groups. (1) group measured volume: consisted of sibelium group, topamax group, single ischemic group, fake operation group and normal rat group. (2) Pathology and HSP?o group: sibelium group, topamax group, single ischemic group, fake operation group. (3)HSP7o dynamic observation group: The reperfusion time point after MCAO were:0h,3h,6h,12h,ld,3d,7d. The MCAO time of all three groups above was two hours, but the reperfusion time was different: the first group was six hours; the second was one day. The rat brains of the group measured volume were dyed with TTC and measured the area by computerized pathological image analyzer. Then the infarct volume and the ratio of infarctvolume were calculated. In HSP70 group, the samples were stained using immunohistochemistric method. The number of positive cells and the stained intensity were measured by computerized pathological image analyzer. 2 control group: the rats of this group were not performed the operation of double renal artery stenosis. The feed time was equal with group one.Results l.The blood pressure of the rats which were operated double renal artery stenosis was high, and four rats suffered stroke. 2.The rats which were performed the operations of MCAO and reperfusion all had right limb parelysis.S.The HSP70 expression of groups using drugs were higher than single ischemic group. 4.The infarct volume of groups using sibelium and topamax were smaller than that of single ischemic group.Conclusions l.The method of double renal stenosis is effective to make RHRSP.2.The improved Longa's method making MCAO and reperfusion model is practicable. 3.Sibelium and topamax can enhance the expression of HSP70; decrease the infarct volume and protect the ischemic cerebral tissue after MCAO and reperfusion. |
