Parti The in vitro culture of human bone marrow stromal cells and Analysis of their phenotype and compenentsAbstract Objective: To culture human bone marrow stromal cells(BMSCs)for long-term,assay theitr phenotype and compenents. MethodsrBMSCs were cultured in vitro by static liquid assay; the phenotype and DNA content were measured by FACS. At the same time,the expression of Vimentin, Desmin, a-Smooth muscle actin and Von willebrand factor in the Child marrow stromal cells were detected by immunochemistry assay. Results: BMSCs could be passaged in vitro for at least nine generations and the levels of CD33 and CD34 on the surface of BMSCs were very low, which suggests hematopoietic cells were absent. Additonally, the expression of CDw90 in child BMSCs was higher than that of adult, demonstrating the child BMSCs are more primitive.The expression of Vimentin declined gradually during passaging process, moreover staining of Desmin, a-Smooth muscle actin and Von willebrand factor were all negative. The results in the present study indicate that the BMSCs in our experiment derived from mesenchymal stem cells and did not contain vascular endothelial cells ;The Culturing Condition displayed influence on growth and differentiation of BMSCs when cultured in Vitro.
