Background: Systemic lupus erythematosus(SLE) is a typical autoimmune disease involved in multiply organs.Monocyte and lymphocyte infiltratiion leads to the damage of multiply organ function.However,the precise molecular mechanism of the recruitment and activation ofmonocyte and lymphocyte remains to be determined.Recently,a new and potent monocyte chemoattractant , MCP-1, has been identified which can chemoattracte and activate monocyte with a high degree of specificity.MCP-1 is one of the chemotactics for lymphocytes and basocytes.These data,together with findings of MCP-1 overexpression in lots of autoimmune disease models,prompted us to explore the possibility that MCP-1 could be implicated in the pathophysiology of systemic lupus erythematosus.Objection: To study the relation between MCP-1 mRNA expression in TECs and WBCs within uric diposit and the activity index of illness for SLE ,to provide new solution for the medical treament of systemic lupus erythematosus.Methods: (T) collecting blood and urine samples from three groups' :SLE, normal and control without autoimmune diseased ヽollecting blood and urine samples of patients with SLE after receiving immunosuppressive agents (3) evaluating the expression levels of MCP-1 mRNA within PBLCsJECs and WBCs by means of quantitative RT-PCR?All patients with SLE fulfilled more than four of the revised criteria for the diagmosis of systemic lupus erythematosus proposed by the American Rheumatism Association(5)The MCP-1 PCR productions were confirmed by southern hybidizationResult: ㏕he level of expression of MCP-1 mRNA was basically low in control:0.42 ?0. 17&0. 47 ?0. 21and normal group:0.31 + 0.14&0.40 ?0. 18(2)The level of expression of MCP-lmRNA in patients with SLE was much higher than that in aboved group which were 0.87 + 0. 23&0. 89 + 0. 15 ㏕he level of MCP-lmRNA expression in PBLCs,TECs and WBCs of patients suffering from SLE was positively correlated to the activity index of illness for SLE,r=0.892(n=25,P<0.01).@The level of expression of MCP-1 in patients with SLE decreased to basic level after the treatment of immunosuppressive agent ,dexamethasone,in 3 weeksConclusion: (D MCP-1 maybe plays an important role in the autoimmune response in patient with SLE(2)The level of MCP-1 mRNA expression in TECs and WBCs within the uric deposit may reflect activity index of illness for SLE.(3)It is guessed that SLE may be treat by some immunosuppressive agents which have an inhibiting effect on the expression of MCP-1 mRNA.
