Objective 1) To observe the morphology and function of chondrocytes seeded onto human Type I collagen sponge of placenta,to find the sponge?s adsorption to chondrocytes and it?s biocompatibility.2) To explore the effect of bFGF and the collagen sponge- chondrocytes complex on the repair of articular cartilage defect through cartilage tissue engineering. Methods 1) Primary and passage chondrocytes were cultured when they were seeded onto the collagen sponge. With light microscope and scanning electron microscope, the chondrocytes morphology and sponge?s adsorption and it?s biocompatibility were observed; Type II collage synthesis were used as markers of histochemistry.2) Chondrocytes were taken from three days old New Zealand unmature rabbits, were cultured in vitro, then were seeded onto the collagen sponge, the sponge- chondrocytes complex were prepared and transplanted into full-thickness defect .The exteral femur condyle defect of left knees were left empty, the media defect were planted with complex, the right knees were not only modeled as left knees ,but also were injected with bPGF. the rabbits were killed at 2~ 4 8. 12.. 1620 weeks postoperation, and the speciments were disected ,macroscope examined histologically, and electronic microscope. Results 1 )Passage chondrocytes seeded onto the sponge maintain the characteristic chondrocyte morphology and Type II collagen phenotype.2) The sponge has excellent adsorption and biocompatibility. 3) There were significant difference in the thickness of repair tissue between the media defect of both knees in 12 weeks after operation, There were no difference in 20 weeks after operation, the typical cartilage tissue was formed and mature chondrocytes were 3 observed under electron microscope after operation. The were no difference in the exteral defect of both knees in 12 and 20 weeks, the left exteral condyle defect were covered with fibrous tissue, but the right ones were covered with both fibrous tissue and cartilage tissue. Conclusion I )Human Type I collagen sponge of placenta can be used as the scaffolds in the three-dimensions culture of chondrocytes.2) The defect of cartilage maybe repaired through cartilage tissue engineering, and the bFGF play an important part in the cartilage tissue engineering.
