| Objective I .To investigate the effect of repairing segmental bony defects with the composite grafting of autologous peripheral blood stem cells(APBSC)and demineralized bone matrix(DBM). 2.To explore the effect of GM-CSF on autologous peripheral blood stem cells(APBSC)mobolization. Methods 1. 20 patients were treated. The bony defects of the patients were resulted from either the resections benign bone tumors, or the operations on case of non-union fractures. The ability of repairing segmental bony defects were observed by serial radiographic and the standard of clinical bone fracture healing. 2. GM-CSF 3ug/kg was injected subcutaneously daily for 3 consecutive days. On the furth day, APBSC harvest was performed with CS-3000 blood cell separator. The peripheral blood, APBSC harvest and centrifugalized APBSC harvest were collected for CD34+ detection, to compare with marrow before peripheral blood stem cell mobolization. Results 16 cases have been followed for 4 to 16 months. All the cases healed primaily except one which was resulted from humerus fracture. Various degrees of osification was found in the cases from 2 to 4 months after having been operated on. 15 cases had union in 6(4-7) months. The osteogenic effect was satisfactory (P<0.05). 2. After adminitration of GM-CSF, the white blood cell count increased significantly, CD34+ cell also increased significantly(P~z0.0 1)0 Conclusion APBSC/DBM is a much effective method for the repairment fo bony defects. |
