Objective To evaluate the effect of human bone morphogenetic protein 2(hBMP-2)through lentivirus combination/Bone MesenchymalStem Cells(BMSCs)/demineralized bone matrix(DBM)on repairing large defect inrabbit femur and to explore thenew treatments for large femoral defects.Method Femoral bone defect models was established by the middle femoral bone cut andsteel plate fixed.Then,animals were randomly devided into 4 groups(n=12 per group): GroupA were not implanted anything,Group B were implanted with DBM.Group C were implanted with hBMP-2/DBM(bone mesenchymal stem cells adhered to demineralized bone matrix).Group D were implanted with LV-hBMP-2/BMSCs/DBM(bone mesenchymal stem cells that were transfected with hBMP-2 through lentivirus combination andadhered to demineralized bone matrix).Three rabbits from each group were sacrificed at2,4,8 and 12 weeks after surgery to evaluate the the repairing effect of femoral defects through HE staining,biomechanical analysisand X-ray examination.Result X-ray revealed A,B,C,D four groups all have different degrees of osteotylus formation,Lane-Sandhu X-ray scores A < B < C < D group(P < 0.05).A,B,C group 2 weeks when there was no statistically significant difference.Biomechanical analysis showed the C,D groups compared with the normal femur,or between the C,D groups had statistical significance(P < 0.05)in 8 weeks and 12 weeks.HE staining in group A showed a little trabecular bonedisordered arrangement in the bone defect area and amount of fibrous tissue;DBM degraded in Group B bone defect area,showed part of the trabecular bonedisordered arrangement and amount of fibrous tissue;Group C bone trabecular in the bone defect area has been basically connect into line,start shaping of cortical bone,recanalization of the medullary cavity was not significant;Group D has basically formed new bone cortex in the bone defect area and existingmedullary cavity with again.Conclusion LV-hBMP-2/BMSCs/DBMcan promote bone conduction and bone induction and osteogenesis after implanted bone defect area.It have good repairing effect on rabbit femoral large defectwith good application prospect.
