Recently some studies have been reported that insulin resistancemay play a role in the pathogenesis of pregnancy induced hypertension.Insulin resistance means the ability of insulin to stimulate the uptake ofglucose impaired and that insulin is unable to exert its biological effectsto normal level. Compensatorily, the bodies are hyperinsulinmic in orderto overcome insulin resistance. Recent investigations show that patientswith pregnancy induced hypertension are hyperinsulinemic and haveimpaired glucose tolerance, and insulin sensitivity indexes decrease inthem. Molecular mechanisms of insulin resistance have been investigated.Point mutations have been identified in the insulin receptor gene insubjects with genetic syndromes of severe insulin resistance, such aspatients with Rabson-Mendenhall syndrome or Leprechaunism or Type AInsulin Resistance.We have screened 29 patients with pregnancy induced hypertension( 10 with mild degree and 19 with moderate or extreme degree ) formutations in exons 17-2 1 of the insulin receptor gene of placenta bypolymerase chain reaction-single stranded conformation polymorphism(PCR-SSCP) analysis. Then PCR products with SSCP variants weresubjected to direct sequencing. We also detected the fasting level ofserum insulin in the patients by radiaoimmunoassay (RIA). Nineteennormal pregnant women were also investigated as controls.Objective 1, To validate if pregnancy induced hypertension isassociated with insulin resistance. 2, To investigate the molecularmechanisms of insulin resistance in pregnancy induced hypertension.3Results1.
