Aim: To prepare the rabbit model of multiple organ dysfunction syndrome(MODS) in early stage after burn To study the effects of Rhodiola(Rho), nitric oxide(NQ), hemoglobin(HB). naloxone(Nal) on the multiple organ dysfunction syndrome(MODS) in early stage after bum in rabbit and analysis its acting mechanism To observe the changes of leucine-enkephalin(L-Enk) content in the arteries of rabbits in early stage after bum. To study the effects of L-Enk in the arteries on the changes in hemodynamics at early stage postburns. Methods: The rabbits were divided into 7 groups, the sham bum group(SB), burn 1 group (B?, burn 2 group (B2), orally taken Rhodiola group (R), burn and Rhodiola therapy group (BR), burn and Nw -nitro-L- arginine therapy group (BL), burn and Naloxone therapy group (BN) All rabbits were anesthetized with ether inhalation. The dorsal skin of rabbits were immersed in 800C (group B? water, 95 (group B2, BR, BL, BN) water, or 370C (SB) water for 30s. This procedure produced a full thickness burn of 30% total body surface area (TBSA) or sham bum. At 48h postburns, all rabbits were anesthetized with intravenous injection of barbiturate (3Omg/kg).Mean arterial pressure (MAP), central venous pressure (CVP), cardiac output (CO) were monitored, and systemic vascular resistance (SVR) and cardiac index (CI) were calculated Pulmonary alveolar perfusion protein (PAPP) and plasma total protein (TP) were determined. The index of pulmonary permeability was calculated from (PAPP/TP)% . Partial pressure of oxygen of arterial blood (PaO,), oxygen saturation of arterial blood (SaC)2) were measured. The levels of serum creatine phosphokinase (CK), glutamic pyruvic transaminase (GPT), urea (BU), creatinine (Cr), blood platelet count (PLT) and I-lB were detected. These data reflected separately the functions of heart, liver, lung, kidney and blood coagulation system. NO; and NO; contents, the metabolic products of nitric oxide in blood and pulmonary alveolar perfusion were measured by Griess method . L-Enk contents in the arteries were measured by Radioimmunoassay (RIA) method. Results: 0 Rabbits of group B2 developed MODS at 48h after burn. The dysfi.tnctions of heart, liver and kidney achieved the criterion of MODS, the lung function declined, SVR increased respectively. ?In comparison with SB . the NO content of group B2 significantly increased in blood and pulmonary alveolar perfusion at 48h postburns (B48h) . ?In comparison with B~?, the functions of heart and kidney were markedly improved, the NO contents in the serum and pulmonary alveolar perfusion markedly raised in group BR at 48h postburns (BR4&h). 0 In comparison with B48? the functions of heart , liver and kidney were not markedly improved, the NO contents m the serum and pulmonary alveolar perfusion significantly decrease in L-NNA therapy group at 48h postburns (BL48? . (~I) In comparison with SB, B48h, BR4~?and BL4$h, I-lB contents in serum markedly raised in group B2, BR and BL at Oh postburns (B0? BROh, BLOh) . NO contents in serum markedly decrease in group B2, BR and BL at Oh postburns (BOh, BR0? BL0?. ?In comparison with B48? CI were markedly improved only in BN at 48h postburns (BN48? . The others were not markedly improved. ?In comparison with SB, the L-Enk contents of mesenteric artery, ranal artery and pulmonary artery were decreased significantly in group B48?.
