Objective To study the expression of CD~ splice variant V6 (CD~v6), transforming growth factor-beta I (TGF- P ~) and the type II transforming growth factor-beta receptor (TGF- P R LI) in normal epthelial cell, fibroadenoma and invasive cancer of the breast and to evaluate their role in tumorgenesis, progression as well as metastasis of breast cancer~ Method The expression of CD~v6, TGF- P and TGF- P R II was determined in 26 case of normal breast tissues, 33 fibroadenoma of breast and 57 breast cancer tissues with immunohistochemical S-P technic. Result The positive rate of CD~v6 immunoreactivity was 3.8% (1/26), 24.2% (8/33) and 70.2% (40/57) in normal breast tissues, fibroadenoma of breast and invasive breast cancer respectively. The positive rate of TGF- P1 was 23% (6/26), 48.5% (16/33) and 73.7% (42/57) in mormal breast gland, fibroadenoma of breast and invasive breast cancer, while the positive rate of TGF- P Rh was 76.90/o (20/26), 39.3% (13/33) and 17.5% (10/57) in normal breast gland, fibroadenoma of breast and invasive breast cancer. Conclusion (1) Overexpression of CD~v6 was closely related to the lymph node involvement ratio of breast cancer. (2) Malignant breast carcinoma cells escaped the growth inhibitory effect of TGF- P ~. Overexpression of TGF- P increased possitivity of the infiltration and metastasis of breast cancer cells. (3) Reduction or loss of expression of TGF- P R II was found in breast carcinomas. Inverse relationship was observed between TGF- P R II expression and the progression of TNM stage. The reduction or loss of TGF- P R II resulted in escape from growth inhibitory effect of TGF- P and metastasis. (4) There 3 was a positive correlation between TGF- P and CD~v6 expression, and the expression of CD~v6 and TGF- P were related to the TN7M classification of breast cancer. Parallel use of the expression of CD~v6 and TGF- P had higher sensitivity and positive predictive value in evaluating lymph node metastasis and prognosis of breast cancer than relying upon only one of them. (5) A negative correlation between the expression of TGF- P and TGF- P R II suggested that malignant progression may be associated with autocrine and paracrine of TGF- P1 in carcinoma cells. Reduction or absence in TGF- P R II expression escaped the growth inhibitory effect of TGF- P ~, So it was helpful to the proliferation, invasion and metastasis of the malignant cells.
