Objective: Endometriosis is defined as endometriotic tissues growing outside the unterine cavity. The cell biological processes responsible for the pathogenesis of this disease are not well understrood. In order to detect the pathogenesis of endometriosis, the cell model was established and the immunohistochemiocal expression of CD54 was evaluated. Methods:1 The eutopic and ectopic endometrium tissues were cultured by filtration andcentrifugation isolated and mixed cell culture.2 The invasiveness of the epithelial and stromal cells of ectopic and eutopicendometrium were exzamined through Trans-well insert plate. 3 For immortalization, the epithelial cell of normal edometrium were transducted an eukaryotic vector expressing SV40 early region gene by Lipofectin or FuGENE6. 4 The immunohistochemical expression of CD54 in endometriotic andendometrium were evaluated using SP immunostaining. Results:1 The success cultured rate of endometrium and endometrotic of centrifugation isolated method were 92.59% and 68.18%, respectively; that of filtration were 84.21% and 61.54%, respectively; that of mixed culture were 100% and 71.43%, respectively.2 The invasive quantity of epithelial and stromal cell in controlsendometrium were 58.14121.52 92.23?1.84, respectively; that in patientswith endometriosis were 90.36?1.78 11264H6.49, respectively; that ofendometriotic were 111.86115.70 142.93+32.63,respectively.3 Three clones were to exist after seven weeks. They grow slowly, soevaluation was not worked. The clones exist seven months. 4 A difference in immunostaining for CD54 was found between EM andNon-EM. Conclusion: 1 Higly purified epithelial and stromal cells cultured from normalendometrium and endometriotic tissues were obtained by filtration andcentrifugation isolated cell culture. The celluar model will be useful for usto investigate the factors that might influence or promote the degree ofinvasiveness of endometriotic cells and contribute to its pathogenesis. 2 The invasion of epithelial and stromal cell from endometrial andendometriotic tissue was differnet. It show that the invasiveness ofendometriotic is important to the pathogenesis of EM. 3 The method was adopted first time in our country to immortalizedepithelial cell of endometrium. 4 CD54 is postulated to constitute a crucial mechanism in the pathogenesisof EM.
