Eliminating Stasis Soup In The Treatment Of Endometriosis Clinical Curative Effect Observation And In Vitro Experiments

Objective:Based on clinical observation,through culturing ectopic stromal cells in endometriosis in-vitro,observes the effects of Xiaoyu decoction on expression of MMP-2,c-Met in human ectopic stromal-cell in endometriosis,discusses it s mechanism of treatment to endometriosis;supplies scientific evidence for clinical application;further enriches etiology and pathogenesis of endometriosis and offeres new ideas and newmethods for clinical treatment.Method:Clinical observation:Sixty EMs patients were devided into the treatment group and the control group at random. The treatment group were mainly treated with Xiaoyu decoction,and the controls were treated with danazol,their symptoms of the treated group and the control group were observed before and after the treatment,CA125and hemorheology indexes were detected before and after the treatment.Experimental research:The eight cases included in the study of pat ients with endometriosis postoperative were diagnosed as endometriosis(ovari orrhexis)by pathologic examination:Specimens from ectopic endometrial tissue during surgeiy,were used to culture in vitro ectopic endometrial cells.After subculture divided into five groups:control group,the group of low-middle-hi gh dosage drugs of Chinese medicine,western medicine group;then added fetal bovine serum,serum containing low-middle-high dosage of Xiaoyu Decoction,serum containing danazol into each group accordingly.Detect the activity of ectopic endometrial stromal cells in each group of drugs by MTT.Inspect th e expression of MMP-2and c-Met of ectopic stromal cell in each group by the method of western blot.All datas are collected to establish a database, statistical analysis,then evaluate the efficacy of Xiaoyu decoction objectively.Results:â‘ The total effective rate of the treatment group and control group was93.3%and83.3%respectively.Both of them could decrease the symptom din-tegra(P<0.01). Com pared with control group had no statistical significance in expression of CA125.Hemorheology indexes had statistical significance before and after the treatment too.(P<0.01).It had statistical significance between the two groups after treatment(P<0.01,P<0.05).That showed the treatment group were beter than control group in improving hemorheology.Liver and kidney function were normal in treatment group after three month,but ALT was increased in three patients in control group.â‘¡the result of MTT:The comparision of serum containing of Xiaoyu Decoction group with fetal bovine serum group after action for twenty-four hours and forty-eight hours,there is a difference in OD(P<0.05).â‘¢The comparision of medicated serum groups with fetal bovine group,there is a difference on expression of MMP-2and c-Met(P<0.05).The comparision of serum containing of Xiaoyu Decoction groups with serum containing danazol group, there is a difference in each of the two groups too.The comparision of serum containing of middle dosage Xiaoyu Decoction group with serum containing danazol group,there is no difference on expression of c-Met(P>0.05)Conclusions:Xiaoyu decoction which based on Yiqiand removi ng bloodstasis method have obvious clinical curative effect,can improve hem orheology indexes,decrease the level of CA125.Serum of Xiaoyu Decoction can suppress the growth of ectopic stromal cells in endometriosis,dosage related, by the growth of time enhances inhibition,and may promote blocking shift of ectopic stromal-cell by reaching down the expressions of MMP-2and c-Met,Consequently, Yiqi and removing bloodstasis method can relief and treat endometriosis by strengthening organism immune system and improving peivic blood circulation.
