Bone morphogenetic proteins (BMPs) are kinds of low molecular weight acidity polypeptides,which can cause stimulation ofundifferentiated mesenchymal cells differentiation into chondroblasts and osteoblasts and then manufacture cartilage and bone.They are members of the transforming growth factor-beta (TGF- p ) superfamily except BMP-1.There are at least 20 fembers of BMPs,among which BMP-2 is considered to be the only one that have the highest osteogenic potential to cause de novo cartilage and bone formation by itself. It is reported that some fembers of BMPs have correlation to occurrence,progress and metastasis of tumors,especially bone tumors. It can be detected rich expression of BMP inosteosarcoma,chondrosarcoma and so on. There is intimate correlation between BMP and malprognosis of osteosarcoma. Expressing or not expressing for BMP in osteosarcoma may lead to the distinctness of clinical features. There is comparatively strong positive expression of BMP in malignant tumors, which have animated osteogenic potential. It is also reported that the total content of BMP in malignant tumors is the 3.4 tunes as-8-it in benign tumors. Bone tumors in oral maxillofacial region are familiar, especially jawbone tumors, which have complicated types. Regarding the correlation between BMP and jawbone tumors, it is devoid of overall systemic investigation at present This study is to aim directly at BMP-2梩he fember of BMPs mat owns the closest relationship to bone tumors as reported.Objectives of this research include: (1) Investigating expression of BMP and its mRNA in common jawbone tumors;(2) Investigating the variation of expression for BMP and its mRNA between jawbone tumors holding hard tissue or holding no hard tissue. (3) Investigating expression for BMP and its mRNA in odontogenic tumors, and analyzing the significance including. (4) Investigating expression for BMP and its mRNA in ameloblastoma, and analyzing the reason why ameloblastoma holds no hard tissue. (5) Investigating expression for BMP and its mRNA in osteosarcoma, and analyzing the relationship between the expression and neoplastic lymphatic metastasis. (6) Cloning human BMP-2 maturation peptide coding region gene fragment and synthesizing human BMP-2 cDNA probe from human normal bone tissue to investigate expression for BMP-2mRNA by in situ hybridization.Materials and Methods1.113 cases of 8 types of jawbone tumors were selected. Information of these cases was collected including age, gender, location and lymphatic metastasis. Among these cases, 70 cases are fresh specimens for studies of extracting total RNA, RT-PCR, in situ hybridization and immunohistochemistry, while other 43 specimens placed on file were selected in order to augmentsamples for studies of in situ hybridization and-9-immunohistochemistry. A normal fresh bone tissue specimen was obtained from a child with condyle splintered fracture to synthesis human BMP-2cDNA probe.2. Using reverse transcription-polymerase chain reaction (RT-PCR), in situ hybridization and immunohistochemistry, we examined the amplification of BMP-2 gene and the expression for BMP-2 and its mRNA in 8 types 113 cases of jawbone tumors.3. Relative content of BMP-2 in cells was estimated by counting average gray value in cells with image analysis system. The results were analyzed by SPSS8.0 for Windows statistical package. The variation of relative content of BMP-2 between jawbone tumors was analyzed by the one- way ANOVA test. The relationship between lymphatic metastasis and expression for BMP-2 and its mRNA in osteosarcoma was analyzed by Chi-square test.Results1. The results of extracting total RNA in fresh specimens. 70 fresh jaw bone tumors specimens and a case of normal fresh jaw bone specimen all manifested three legible characteristic electrophoresis strips of total RNA: 5.8S,18S,28S.There was no obvious degradation and no genomeDNA.The A260/A280 was 1.75 ?0.12 and the concentration of total RNA was 216.8 ?10.2 M g/ml.The results meaned that the quality of t...
