Objective To investigate the relationship between Osteopontin (OPN), integrinα3,β1 subunit expression and the occurrence and metastasis in colorectal cancer and the clinical significance. Methods S-P immunohistochemistry and situ hybridization, was used to determine the expression of OPN and mRNA to evaluate their effects on tumorigenesis and progression in 60 tissues of colorectal cancer: 32 of which had metastased to lymph nodes , 28 cases no metastasis. 12 cases adenoma, 13 cases normal mucosa. OPN expression was detected by Westernblot by the clinical fresh sample : 8 cases moderate differational colorectal carcinoma , 2 cases metastastic lymph node colorectal carcinoma and 1 case adenoma. S-P immunohistochemistry was used to determine the expression of integrin a3 andβ1 subunit in the same 60 samples. Results The rate of OPN expression in colorectal cancers was 80.0% (48/60), metastasis tumor was 84.3% (27/32), nonmetastasis tumor was 75.0% (21/28) ,adenoma was 91.6%(ll/12),but no expression was found in normal mucosa. OPN expression level had a correlation with the differentiation of tumor , lymph node metastasis and Dukes'stage (P <0.05) ;but had no correlation with the patients'...
