Objective : Exploring the in-vitro cytotoxic assay of horse anti-ovarian-cancer-antibody and investigating the distribution of 131I-labeled anti-ovarian-cancer antibody in nude mice with transplanted ovarian cancer.Methods: Cytotoxicity test mediated with complement and MTT test is conducted to measure the liter of horse antiserum against ovarian cancer and its cytotoxicity to target/non-target cancerous cell. Ammonium sulfate precipitation is applied to purify and extract anti-ovarian-cancer-antibody (IgG). SPECT and isotope measurer are used to detect the distribution of 131I-labeled anti-ovarian-cancer antibody in the nude mice and the radiation in mice's organs for analysis.Result: With complement, the purified Anti-ovarian-cancer horse immunoglobulin has showed stronger cytotoxicity (killing rate) in high concentration compared to median concentration or low concentration (p<0.05). The cytotoxicity of horse anti-ovarian-cancer antibody to the target/non-target cell indicated the killing power's dependency upon concentration and time (p<0.05~0.01), which is valuable for statistical analysis.The research result indicates, in the experiment of 131 iodine-labeled anti-ovarian-cancer horse antibody distributed in tumor transplantednude mice, the distribution of labeled antibody is defined by time and space, the radiation value is measured in the tumor tissue and major organs, and the sense to radiation is evaluated with SPECT for tumor mass and the whole body. It was observed that radiation is apparently stronger in nude mice's transplanted tumor mass than in some organs and tissue (p<0.01). Image showed close correlation between the definitions of intraperitoneal administration and the T/N ratio, i.e. the ratio of labeled antibody content within tumor mass and that within normal organ and blood. Measurement indicated absolute value of radiation intensity in some organs was similar to that in the tumor mass. 8 days aften applied does,the cancerous tissue appear piece meal necroses in naked eyes and infra-microscop,the ovarian cancerous transplanted mice weren't observed to have any obvious intoxication and other side-effects.Conclusion: This research indicates that anti-ovarian-cancer horse antibody has efficacy in killing target/non-target cancerous cell to some extents, which can be regarded remarkable for treatment to ovarian cancer metastasis be of worth for further clinical trial,That 13 liodine-labeled anti-ovarian-cancer horse immunoglobulin has assembled for long in the transplanted ovarian cancer mass of nude mice, showing potential for clinical application though this antibody still needs further improvement and purification.
