Kindling which would not cause focus in brain is thought to be an ideal animal model to study human chronic epilepsy, because it is similar to human epilepsy in electroencephalogram and behavior.Succinic acid , an ubiquitous natural activity, is an important content in amber which is a traditional antiepileptic drug. Succinic acid plays inhibitory effect on central nervous system and its anticonvulsant effect had been reported, yet its antiepileptic effect on animal kindling model was not reported. We investigated the antileptic effect of succinic acid on PTZ(pentylenetetrazol) kindling and amygdala kindling models in rats. The follows are the results:1.PTZ chemical kindling and amygdala electrical kindling models in rats were established successfully.2.Succinic acid 100-400mg-kg-1 ip dose-dependently inhibited PTZ kindled seizure and decreased Ono's stage and percentage of Ono's Stage 6seizure(P<0.01).3. Succnic acid 100-400mg-kg-1 ip dose-dependently inhibited amygdala kindled seizure in rats, elevated the afterdischarge threshold and reduced the Racine's stage and the percentage of Racine's Stage V seizure (P<0.01).4. Succinic acid 200-400mg-kg-1 ip significantly prolonged the latancy of clonic and tonic phase of picrotoxin-induced convulsion in mice (P<0.01).The results suggested that succinic acid inhibited seizures of the PTZ kindling and amygada kindling. Succinic acid prolonged the latency of mice caused by picrotoxin, a GABAA receptor antagonist. This indicated that succinic acid can block the inhibitory effect caused by picrotoxin, its mechanism is probably related to blocking the GABAA receptor and enhance GABAergic system.
