Objective To study the relationship between tumor suppressor gene PTEN,P16,P15 and primary cervical carcinomas. Methods By use of immunohistochemistry,the protein of PTEN,P16 and P15 were detected in 20 normal cervical tissues and 71 primary cervical carcinomas including 62 cervical squamous carcinomas and 9 cervical adenocarcinomas Results The loss expression rate of PTEN protein was 5% in normal cervical tissues, the rate of P16 protein was 10%and all samples of normal tissues were found to express P15 protein. The total rate of PTEN protein expression was 33.80% in the 71 cervical carcinomas. The rate of loss expression of PTEN protein in cervical squamous carcinomas and cervical adenocarcinomas were 37.10%and 11.11%. Gene PTEN was associated with squamous carcinomas, but not related to cervical adenocarcinomas , and the expression of PTEN protein was not irrelevant to the stage, grade and metastasis of lymph nodes. The total rate of P16 protein expression was 11.26% in the 71 cervical carcinomas. The loss expression rate of P16 protein in cervical squamous carcinomas and cervical adenocarcinomas were 8.06% and 22.22% The loss of P16 protein was not associated with cervical carcinomas ,but the high expression of P16 was found in the samples of cervical squamous carcinomas, the rate was 56. 45%, which not irrelevant to the stage grade and metastasis of lymph nodeso (4)The total rate of P15 protein expression was 9.86% in the 71 cervical carcinomas. The loss and high expressionof P15 protein were not associated with cervical carcinomas. Conclusions In the tumor development or progression of primary cervical carcinomal,gene PTEN is a important role in cervical squamous carcinoma and not in cervical adenocarcinomas. There are no relationship between the loss of protein of gene P16 or P15 and primary cervical carcinomas.lt is possible that the high expression of P16 protein is a symptom of high-risk HPVs in primary cervical carcinomas. The loss of p16 in the adenocarcinomas was much higher than the squamous carcinoma, it was mean that the loss of p16 was more important in the adenocarcinomas .Measurement of the expression of PTEN and P16 protein in patients'tumors might help in predicting prognosis .directing clinical treatment and providing theories for genetic therapy.
