| Diabetes Neuropathy (DN) is one of the most popular complications of Diabetes, is common neurodegenerative diseases in aged people, imparing the health of patients and taking heavy burden to society, familites and patients. Due to the pathnogeny and pathnogenesis haven't very clear yet, there aren't specific drugs to the treatment of DN by now. In recent years, some clinical curative effects have been achieved with traditional Chinese medicine (TCM) therapy on DN.But, current rereach are focus on peripharal neuropathy and clinical study, the reports on central neuropathy, particularlly about the mechanisim are very rare.Shen Xian Tang decoction (SXTD), developed according to TCM theories and modern pharmacological research achievements, is a pure compound recipe of Chinese herbs with effects of "benefiting essence and nourishing marrow, activiting blood criculation and dredging the obstruction of meridians, strengthening the brain and replennishing resourcefulness".Precious experiments showed the Shen Xian Tang decoction could distinctly ameliorate hearing and memory ruction in brain aging mice with carbohydrate metabolic disturbance induced by D-galatose, and multi-infarct rats modeled by injecting liquid paraffin into the left vertricle. SXTD can prevent the nerve cell from the pathologic changes of degeneration or mecrosis in nurve cells caused by free radicals.The purpose of the present study in the discuss the pathnogenesis of diabetes neuropathy in DN mice model caused by STZ injection, and the pharmacological mechanism of Shen Xian Tang decoction. Firstly, we successfully cultured the nerve cells of rats' hippocampi; In order to evaluatate the reaction of nerve cells on high glucose inbubation and the protective effects of Shen Xian Tang decoction,such indexes as the livability of nurve cells, cytosolic calcium and mitochondrial membrane potential were observed; To evaluate the effects of SXTD on DN mice model abducted by STZ, such indexs as blood glucose, obtaining orientation memory in water maze,expression of neurotrophic factors in hippocampi of brain were observed.The results showed:1. The environment of high glucose has damnification to nerve cells;2. Shen Xian Tang decoction have the protective effect on the damnification to nerve cell caused by high glucose, reducing the density of basal cytosolic calcium, increasing mitochondrial membrane potential of nerve cell;3. Shen Xian Tang decoction improved significantly the memory of the STZ models. Model animals given SXTD carried out Morris water maze in shorter time and shorter distance with fewer errors and highter obtaining ratio than those which were not given SXTD; Carried out obscura in fewer enter times and longer latent period than those which were not given STXD.4. Shen Xian Tang decoction can reduce the blood glucose of STZ mice models somehow;Increase distinctly the expression of NGF, BDNF, NT-3 in hippocampi.Conclusion:We can produce animal model which can mimic the changes in behavior and biochemistry N neurophysiology of DN by using high glucose(16.7mmol/l)and STZ(0.2ml/10g).Shen Xian Tang decoction have entensive protective effects in nerve cell models caused by high glucose inbubation and STZ DN animal models and Shen Xian Tang decoction may has favorable application prospect on prevention and treatment of DN. |
