Reverse transcription-PCR (RT-PCR) were applied to detect mRNA expression of cancer cells in the blood of cancer patients,which might forecasted the prognosis and monitored the efficacy of therapies.To a certain extent,Carcinoembryonic antigen(CEA) was expressed in colorectaL,gastric,and pancreatic tumor,breast, non-small cell lung,and head and neck carcinoma. That CEA was shed into serum is useful as a tumor marker to be detected in about 50% of colorectal cancer patients . RT-PCR was used to detect expression of CEA mRNA in blood and to analysis the clinic significance in the experimentObjectiveTo detect expression of CEA mRNA and CK19mRNA in blood and to analysis the clinic significance in colorectal cancer.Methods and materialsThe patients with colorectal cancers from 2000 to 2001 who had been received surgical therapies.CEA level of peripheral blood?CEA mRNA and CK19rnRNA expressions in peripheral blood and meserrteric vein blood were detected by RIA(radioimmunoassay) and RT-PCR in by RIA (radioimmunoassay) and RT-PCR in patients with colorectal cancers. According to Dukes stage,the patients were divided into Dukes A (20), B (25), C (17), D (9). Other factors such as different kinds of pathological or the site of the tumor were also taken into account. Among 9 patients with Dukes D stage,metastasis of lung (lcase),peritoneum(lcase),and liver(7cases) were discovered.CEAs __Positive is determined above Sng/ml. The production of RT-PCR was analyzed by agarose gel eletrophoresis. CEA mRNA and CK19 mRNA molecular weight were standardized by markers in the positions of 199bp and 200 bp respectively. X2 analysis was used as the statistics method in this protocolResultsThe positive expressive rate of CEA mRNA and CK19 mRNA in mesenteric vein blood is 82.9%(29/35)and 71.4%(25/35),which was higher than expressive rate of 69.1%(49/71) and 59.2%(42/71) in peripheral blood in patients with colorectal cancer. That CEA level exceeded 5 ng/ml in peripheral blood was about 45. l%(32/71).The positive rate of CEA, CEA mRNA and CK19 mRNA in patients with Dukes C+D stage colorectal cancer was higher than those with Dukes A+B patients.The positive rate of CEA mRNA and CK19 mRNA with liver metastsisis was high with 100% and 85.7% respectively. To detect CEA, CEA mRNA and CK19 mRNA in peripheral blood were associated to increase the positive rate in corlorectal cancer.Expression of CEA mRNA and CK19 mRNA in peripheral blood was negtive in 20 patientswith benign disease.Conclusion1. The detection of CEAmRNA and CK19mRNA expression in mesenteric blood during the operation and peripheral blood could be better in reflecting the development of the disease and in evaluating the patients' prognosis than CEA could.2. The detection of CEAmRNA and CK19mRNA expression in mesenteric blood is great significant in early discovery of liver micrometastasises in colorectal cancer.3. The combined detecting of CEA, CEAmRNA and CK19mRNA expression can increase the sensitiveness of screening colorectal cancer.
