Renal carcinoma is one of the common malignant tumors in urinary system.Its incidence ranks second among all urinary tumors. It's widely recognized that oncogenesis of kidney carcinoma is a multigene , multistage process, which concerning the change of many genes, including the protein governing DNA repair, signal transduction and cell cycle controLAnd the cell cycle is the final pathway of cell proliferation and differentiation. It has been proved that the occurrence of oncogenesis is in close relationship with the oncogene activation, the tumor suppressor gene inactivation and the disorder of cell cycle modulation.In 1994, Polyak found a heat-stable protein of 27KD and named it p27kipl.It can inhibit the CDK activity , inactivate the procedure Gi-S stage transduction and therefore stop the cell cycle in the GI stage via binding with the CDK or the CDK-cyclin compound.Tumor suppressor gene and oncogenesis of kidney carcinoma are enjoying great popularity in the world nowadays, while the report of the relationship between p27 gene and ocongenesis of kidney carcinoma is rare.In this report, we investigate the relationship between amplification and expression of p27 gene and the oncogenesis and developing of renal carcinoma , and explore the effect of p27 gene transfection on the renal carcinoma cell line GRC-1 with the employment of immunohistochemitry , cell culture and gene-4-transfection tecthnique.The results are as follows:In the 86 speciments of renal carcinoma and paracarcinoma, p27 protein expression is detected.The result showed that the positive expression rate of p27kipl protein is 44.2% (38/86) in tumor (clear cell carcinoma 43 cases, granular cell carcinoma 22, mixed cell carcinoma 21) and 74.4% (64/86) in paracarcinoma.The diffence of p27 protein expression between them was significant (P<0.05) ?I stage 20 cases, II stage 18 cases, IIIstage41 cases, IVstage 7 cases, and the positive expression rate was 75.0% (15/20), 44.4% (8/18), 34.1% (14/41), 14.3% (1/7) respectively.So, there is relationship between p27 expression and the stage(P<0.05).Meanwhile, I grade 44, II grade 19, III grade 23, andp27 positive rate are 56.8%, 42.1% and 21.7% respectively.So, there is a negative correlation between p27 expression and grading(P<0.05).Transfect the p27cDNA into the renal cell carcinoma cell line GRC-1, and investivgate its effect on the cell culture, apoptosis and cell cycle, and compare it with the contrast group.lt reveals that the overexpression of p27 may inhibit cell proliferation, induce cell apoptosis, and stop the cells in GI stage.In 24 hour, we can observe p27-triggered apoptosis, which peaks at 72 hour.So it's of great value searching p27 gene therapy utilizing suitable intermediary.Our study shows the correlation between underexpression of p27 and the oncogenesis, progression and prognosis of p27.The expression of p27 protein is a reliable marker for determining its prognosis. p27cDNA gene transfection may become a new way for renal cell carcinoma treating.
