Objective: To determine whether the peripheral blood mononuclear cells of hemodialysis uremia patients are activated to increase interleukin-8 mRNA expression with the technique of molecular biology. To provide theoretic basis for the clinical diagnosis and treatment of uremia.Methods: To label and detect interleukin-8 cDNA probe with DIG. Select 15 hemodialysis uremia patients and 15 normal persons to divide into three groups which were prehemodialysis group >, posthemodialysis group and normal group. Obtain peripheral blood momonuclear cells (PBMC) by Ficoll density gradient, which were detected interleukin-8 mRNA by situ hybridization and dot hybridization. All data were made statistics analysis after mapping analysis.Results: There wasn't expression of interleukin-8 mRNA in normal group,while there were notable expression of interleukin-8 mRNA in prehemodialysis and posthemodialysis groups,of which the rate of positive expression were 91.4% and 90.2% respectively. Between the two groups, discrepancy didn't have statistics meaning ( P>0.05 ) . Mapping analysis result showed: Compare with the normal group, the value of optical density in the other two groups increased apparently (P <0.01 ) , but between the two groups, discrepancy didn't change ( P>0.05 ) .The dot hybridization showed the same result.Conclusion: There wasn't interleukin-8 mRNA expression in normal group.while there were notable interleukin-8 mRNAexpression in prehemodialysis and posthemodialysis groups. The results showed that the uremia peripheral blood mononuclear cells were activated and had no matter with the time of hemodialysis and the exclusion of exotoxin.
