ObjectiverTo observe the thrombolysis and anti-thrombosis effects of MPAP;To study the actions of MPAP on blood coagulation,platelet aggregation and fibrinolytic activity.Methods: 1 .Five groups in vitro were designed in order to observe if a plasma clot formed or the plasma clot dissolutime.2.Two kinds of fibrin plates (plasminogen-free fibrin plate and plasminogen-rich fibrin plate) were made to estimate the activity of MPAP.Five different concentrations of MPAP solution were dropped into each hole of the two kinds of fibrin plates.The diameter of the clear zone was measured after 16 hours.B.The thrombus models in the moral artery in rabbits were established by injuring the tunica intima and injecting thrombasein;The thrombus in the artery was observed for 2 hours by cardio angniography after MPAP was administered;The TT,PT,KPTT and the content of fibrinogen of rabbits were assayed 20 minutes post-MPAP.4.The thrombus models of pulmonary in rabbits were established by forming a thrombus in the jugular vein and let it block the pulmonary artery;The levels of t-PA and PAI were assayed 20 minutes post-MPAP;The embolus taken out from pulmonary artery was observed with SEM.S.The obstruction time (OT) in the model of carotid thrombosis induced by electrial stimulation in rats post-MPAP was measured;ELT was assayed 20 minutes post-MPAP.6.The action of MPAP on platelet aggregation was observed in rats 20 minutes post-MPAP.Results: 1 .The clot dissolutime was similarCP>0.05 )in both MPAP plasminogen-free and plasminogen-rich groups in vitro;The clot couldn't form if MPAP and fibrinogen were incubated at 37 for 3 minutes before thrombase was added into the mixture.2.The same concentration of MPAP had similar clear zone in the two different fibrin plates;Compared with UK,the activity of MPAP was 61.35IU/mg.3.Compared with themodel group,the successful rate of thrombolysis was higher in MPAP group ( P< 0.01 ) ;The TT,PT,KPTT were shortened and the content of fibrinogen decreased (P< 0.01) ;The correcting test of PT and KPTT showed that the levels of factor II and IX decreased post-MPAP.4.Compared with the model group,the wet weight of embolus in lung artery decreased (P<0.01) ;The filament of fibrin of medicated group became thin and short.5.Compared with the model group,OT was prolonged (P<0.05) ,and ELT was shortened ( P<0.01 ) in MPAP group.6.Compared with the model group,platelet aggregation induced by ADP was impressed in MPAP group.Conclusion: l.MPAP showed strongly fibrinolytic activity which did not rely on stimulating plasminogen.2.MPAP possessed actions on anti-thrombosis and thrombolysis.3.MPAP promoted the fibrinolytic activity and inhibited the blood coagulation and platelet aggregation.
