The Effect On Cell Apoptosis And The Mechanism Of Basic Fibroblast Growth Factor In Rat Retinal Ischemia-reperfusion Injury

Objective To observe the expression of cell apoptosis and correlative genes c-fos/c-jun gene in rat retinal ischemia-reperfusion injury,and to study the effect of basic fibroblast growth factor (bFGF) on them for investigating the mechanism of bFGF.Methods 35Wistar rats were divided into ten groups randomly. Rasing intraocluar pressure to induce retinal ischemia.One hour later .reducing the pressure to normal level and the blood stream recovered.lt led to the retinal ischemia-reperfusion injury.Some rats were treated with bFGF.The expression of c-jun/c-fos gene were determined by immunal histochemistryjand the apoptosis were determined by TUNEL(In Situ Cell Apoptosis Detetion Kit I, POD).The positive stain sections were analysed by computer image analysis system.Results There were no positive cells of TUNEL,c-jun and c-fos gene in normal retina and retinal outer nuclear layer(ONL);TUNEL positive cells were found in retinal ganglion cell layers (RGCL)and retinal inner nuclear layers (INL) at each group without bFGF,peaked at 24hours,but the amount at bFGF-treated groups 8,12,24,48hours significantly decreased(p<0.05);the c-jun and c-fos positive cells were firstly found at 1 hours in RGCL and INL,c-jun was peaked at 12 hours ,c-fos was peaked at 8 hours ,then the amounts decreased.No positive cells were found at 48 hours group.The amounts of c-jun ,c-fos positive cells in groups with bFGF-treated at 4,8,12,24hours decreased (p<0.05).Conclusion l.The expression of apoptosis,c-jun gene and c-fos gene can be found in retinal ischemia-reperfusion injury;2.bFGF caninhibit the expression of c-jun gene and c-fos gene furthermore it also can prevent cells from apoptosis;3.In retinal ischemia-reperfusion injury c-jun and c-fos gene probably induce cell apoptosis .Postgraduate:Gao Yunxia(Ophthalmology) Directed by Prof.Niu Yingjun...
