Objective 1. To find out the effects of basic fibroblast growth factor(bFGF) and recombinant human bone morphogenetic protein(rhBMP) by acting individually or jointly on the proliferation of human periodontal ligament cells(PDLCs) and select their optimal density. 2. To study the effects of bFGF and rhBMP by acting individually or jointly on collagenase( IK IV) levels of PDLCs, and to explore their internal correlations and select optimal density.Methods 1. To select the first premolars extracted due to orthognathous treatment need and scrape the one-third PDL tissues of fang's intermediate to be samples, PDLCs culture technique,MTT assay and enzyme kinetics methods were used to observe dynamically the effects of gradient contents of bFGF and rhBMP by acting individually or jointly on the proliferation of PDLCs. 2. Immunohistochemistry method was used to observe dynamically the effects of gradient contents of bFGF and rhBMP by acting individually or jointly on the collagenase( IK IV)levels of human PDLCs.Results 1. The individual action of bFGF or rhBMP can promote the proliferation of human PDLCs(P<0.01); Significantly greater proliferation was found in bFGF(10)+rhBMP(200) (sum of the best density) compared with individual action(P<0.01); bFGF(10)+rhBMP(200) was observed dynamically to act on PDLCs from 1 to 7 days and to indicate continuously increasing promoting proliferation trend(P<0.01). 2.The results of immunohistochemistry were used to conduct quantitative analysis to show: in bFGF(10) group, the expression of CollagenaselV was significantly inhibited(P<0.01); in rhBMP(200) group, the expression of Collagenase IV was induced(P<0.05) ; in bFGF(10)or rhBMP(25) group, the expression of Collagenase II was significantly induced (P<0.01) ; inhibitive effects of bFGF(10)+rhBMP(200) on levels of Collagenase( IK IV) were observed dynamically to appear continuously increasing trend within 7 days(P<0.05). 3. There was a significantly negative correlation between rhBMP and the levels of human PDLCs Collagenase II (PO.01); There was a significantly positive correlation between rhBMP and the levels of human PDLCs CollagenaseIV(P<0.01).Conclusions Individual action of bFGF or rhBMP can promote the proliferation of human PDLCs, and combined action of bFGF and rhBMP can significantly promote the proliferation ofhuman PDLCs and inhibit the expression of Collagenase( IK IV), and they had synergistic action. bFGF (10) +rhBMP (200) was optimal density.Zhao zheng (Oral clinical medicine) Directed by Zhang guang yun...
