Objective: To examine the effects of mifepristone combined with anordrin or famoxifen on the endocrine function of chorion and decidua and on levels of TNF- a and IL-2 of decidua in early human pregnancy in vitro for investigating the mechanisms of antigestational action of mifepristone combined with anordrin or tamoxifen.Methods: The project was studied by the methods of cells culture and RIA.Results: 1. In the culture medium of chorionic tissue treated with 1000 nM mifepristone and different concentrations of anordrin (100 nM,200 nM ,400 nM ,800 nM) for 48h, levels of HCG P E2 and 6-keto-PGF1 were decreased significantly (P<0.01, P<0.05) while the level of TXB2 was increased significantly (P<0.01) when compared with the control group. The ratio of 6-keto-PGF/TBX2 was declined greatly (P0.01). In the culture medium of decidual tissue treated with the same method the level of E2 was decreased (P<0.05), levels of TNF- a and IL-2 were increased (P0.01, P<0.05),the level of TXB2 was increased in the culture medium containing 1000nM mifepristone and 400nM or 800nM anordrin, when compared with the controlgroup.LevelsofHCG P ,TXB2, IL-2,TNF-a in culture medium of chorionic and/or decidual tissue related with the dose of anordrin. 2. In the culture medium of chorionic tissue treated with 1000nM mifepristone and different concentrations of tamoxifen for 48h, levels of HCG P E2 and 6-keto-PGF were decreased significantly (P<0.05, P<0.01) while the level of TXB2 increased significantly (P<0.05) when compared with the control group.The ratio of 6-keto-PGF/TXB2 declined significantly (P0.01). In the culture medium of decidual tissue treated with the same method, levels of E2^ 6-keto-PGF PRL were decreased (P0.01, P0.05) while the level of TNF- a was increased significantly(P<0.01) when compared with the control group. Levels of E2 6-keto-PGF TNF-a in culture medium of chocionic and /or decidual tissue were related with the dose of tamoxifen. 3. Effects of mifepristone combined with anordrin on levels of TNF-a , IL-2 TXB2 from decidual tissue(P<0.01, P<0.05, compared with control group) were greater than mifepristone administed alone (no differerce when compared with the control group,P>0.05). Effects of mifepristone combined with tamoxifen on levels of E2 TNF-a from decidual tissue were greater than mifepristone administed alone (P<0.05, P0.01) and effect on 6-keto-PGF level (P0.05, P0.01, compared with the control group) was greater than mifepristone administed alone (no difference when compared with the control group, P>0.05).Conclusion: Mifeprstone combined with anordrin or tamoxifen affects the endocrine funtion of chorionic and decidul tissue significantly, and changes production of IL-2 TNF- a from decidual tissue. Effects of mifepristone combined with anordrin or tamoxifen on endocine function of chorionic and decidual tissue and on production of cytokine relate with the dose of anordrin or tamoxifen. Effects of mifepristone combined with anordrin or tamoxifen on endocine function of chorionic and decidual tissue and on production of cytokine were greater than mifepristone administed alone.
