BackgroundNeuropeptide Y(NPY) is a kind of polypeptide with 36 amino acid residues, and usually coexists with norepinephrine(NE) in teleneuron. Its wide bioactivity, especially its strong regulation on cardiovascular system has been recognized. Since 1990s the application of angiotensin II(AII) antipeptide has been an important development of the study on renin-angiotensin system(RAS), and now it has become the hot spot of the study on antihypertensive drugs.ObjectiveBy observing plasma NPY levels in patients with hypertension, their significance in the pathophysiologic process of hypertensive development has been explored. And by observing changes of NPY levels after losartan treatment, the correlation of All antipeptide with NPY has been further explored. MethodsThe experiment includes two steps. In the first step, plasma NPY levels in 80 patients with hypertension were measured by RIA and compared with 40 normal persons as control group. In the second step, 80 patient with hypertension were randomly divided into two groups. 40 patients were treated with losartan 50-100mg per day for 4 weeks, and the other 40 patients were treated with metoprolol 25-50mg per day for 4 weeks, then their plasma NPY levels were measured again and compared with that before treatment. All were also measured by RIA in two groups before and after treatment. All the data were analyzed statistically.Results1 The plasma NPY contents in 80 patients with hypertension and 40 normal persons were 148.4+45.2 pg/ml and 118.5+30.5 pg/ml respectively.The difference between two groups was significant (p<0.01).2 The levels of plasma NPY in patients with hypertension were significantly higher. The higher of blood pressure, the higher of NPY level. The NPY contents in patients with III grade hypertension(n=24), II grade hypertension(n=28) and I grade hypertension(n=17), were 162.4+42.7pg/ml, 148.5+41.Ipg/ml, 137.3+32.6pg/ml respectively. The NPY content in patients with hypertension whose blood pressure was normal at present was 117.3+22.6 pg/ml, without difference to thatn in control group(p>0.05).3 The hypertension group was compared with the control group according to the absolute level of risk factors of cardiovascular disease in 1999WHO/ISH of Hypertension Guidelines for the Management of Hypertension. The plasma NPY contents in patients with hypertension polari-high risk group(n=ll), high risk group (n=24) and middle risk group(n=45) were 157.4+16.8pg/ml, 146.5+18.3pg/ml, 135.3+17.6pg/ml respectively, with significant difference to that in control group (p<0.01). The results showed that the more of the risk factors of cardiovascular disease, the higher of the plasma NPY.4 The average systolic pressures before and after losartan treatment in 40 patients with hypertension were 175.2+25.6 mmHg and 135.5+11.2 mmHg, the average diastolic pressures were 103.4+12.3 mmHg and 83.7+8.2 mmHg respectively. The average systolic pressures before and after metoprolol treatment in the other 40 patients with hypertension were 180.5+21.6 mmHg and 120.4+10.3mmHg, the average diastolic pressures were 110.3+11.9 mmHg and 78.6+8.0 mmHg respectively. It was clear that the blood pressures lowered significantly after losartan or metoprolol treatment for 4 weeks in the patients with hypertension(p<0.01), and the decompression effect of metoprolol was relatively better than losartan.5 The contents of plasma NPY before and after losartan treatment were 152.4+12.3pg/ml and 125.5+9.5pg/ml respectively, with significant difference(p<0.01). But the content of plasma All increased after losartantreatment. It was 49.6+7.1pg/ml before treatment, and 62.4+7.8pg/ml after treatment. There was significant difference too(p<0.01). The content of plasma NPY or All after metoprolol treatment had no notable changes. NPY contents were 144.1+11.3pg/ml and 140.6+12.2pg/ml, All contents were 45.5+5.8pg/ml and 48.3+7.2pg/ml respectively, without statistical difference(p>0.05).
