ObjectiveNasal polyps is a kind of clinically common disease, It's incidence in adults is about l%-2%, the cause is still not clear. Recent research indicates that eosinophils infiltration is the most characteristic pathological change of nasal polyps. Bachert etc pointed out that the mechanism of eosinophils infiltration mechanism into nasal polyps is an important point in the first place for us to understand its cause. Although single cytokine change in nasal polyps has been reported, but in view of the effect of cytokines which usually possesses the cross-domain communication characteristic, therefore, study on cytokine network interaction has provided a more value on pathogenesis of nasal polyps; Which invite us to investigate the expression of a set of cytokines, such as IFN- IL-2, IL-, IL-5, IL-10 and GM-CSF , and eosinophils infiltration in nasal polyps, we can then study it's effect on nasal polyps.MethodThe concentration of IFN -Y. IL-2, IL-4, IL-5, IL-10 , GM-CSF was quantified by enzyme linked immunosorbent assay (ELISA) in the total of 30 cases (no case history of asthma, no allergic rhinitis and aspirin intolerance), and the number of eosinophils was counted in the same tissues under microscopes. The inferior turbinate mucosa sample of10 surgical patients (no allergic rhinitis, no chronicrhinitis and chronic sinusitis) was taken as the control group.ResultsThe statistical analysis showed that the levels of IL-4, IL-5 and GM-CSF were significant difference (P<0.05) between the nasal polyps group and the control group. The concentration of IL-4 IL-5, GM-CSF in nasal polyps was 60.4 ± 10.8pg/mg , 84.6 ± 15.7pg/mg , 45.7 ± 6.8pg/mg, respectively, and the concentration of the inferior turbinates was 18.3 ± 3.1pg/mg , 12.9 ± 3.9pg/mg , 19.0 ± 4.4pg/mg respectively ; The concentration of IFN- Y , IL-2 in nasal polyps was 25.1 ± 8.9pg/mg 7.8 ± 1.7pg/mg respectively, while the concentration of inferior turbinates mucosa was 14.6 ± 4.0pg/n, 7.5 ± 1.9pg/mg respectively, the statistical analysis showed that the production of IFN- Y and IL-2 have no significant difference (P , 0.05) between the nasal polyps group and the control group. There is a positive correlation between the concentration of IL-5 GM-CSF and the extent of eosinophils infiltration (the former r=0.891, the latter r=0.704; P<0.05).ConclusionThis study demonstrated that there existed an inductive compartments, namely the microenviroment in which immune-effector and inflammatory cell function is modulated in nasal polyps. The local production of IL-5 and GM-CSF may play a key role on the eosinophil.infiltration and accumulation; The upregulation of Th2-typed cytokines such as IL-4,IL-5,IL-10 may enhance the process of pathophysiology of nasal polyps and Th2-typed cytokines can be interfered as a target molecule for new treatment of this disease.
