[objective] To establish the gene expression profiles associated with the malignant progression of glioma which can help us to understand the molecular mechanism of the development and the progression of glioma, as well as the gene therapy of the glioma.[Method] To Collect primary tumor specimen , tumor specimens of two recurrence and the adjacent normal brain tissue of an individual patient. Analyzing the gene expression patterns of above four tissues based in cDNA array. Drawing the rule of differentially expressed genes by retrieve in the GeneBank.[Result] We collected tumor specimens of a same individual patient with a transformation from ganglioglioma, then to anaplastic ganglioglioma, finally to glioblastoma diagnosed by morphological pathology and there were no cancerous cells in the adjacent normal brain tissue. Based comparing with the signals of normal brain tissue, there were 403,451, and 386 differentially expressed genes more than 2 fold of specimen ? , ? ,?, respectively. There were 46 genes always differentially expressed in different grade which maybe the special gene expression pattern of the tumor. Followed with random matching between four tissues, we found 199 differentially expressedgenes more than 3 fold which maybe the gene expression profiles associated with malignant progression of glioma. By retriving in GeneBank we believe that at initial phase of brain tumor, changes of immune-recognize genes andimmune-inhibition genes were co-existed, as well as the apoptosis-inhibition of tumor cell, and impair of DNA-repair gene. In anaplastic phase, oncogenes were stimulated, suppressor oncogene were inhibited, and some genes associated tumor angiogenesis, metastasis were up-regulated. There were significantly up-regulated genes associated with mitochondria fuction. In GBM phase, stimulation of oncogene and inhibition of suppressor oncogene could still be observed; more genes associated with high invasive ability of tumor were up regulated.[Conclusion ] (1) The gene expression patterns of different grade braintumor with malignant progression in same individual patient can help us to further study the regulation of genes, the molecular pathology and molecular mechanism of the development of glioma. (2) By GeneBank datebase, it's the first time we found that the new function of Pim2 and EHM2 genes which play a significant role in the malignant progression of glioma.
