[Objective] To study the molecular mechanisms of malignant progression of glioma, and detect the expression pattern of DNA repair genes in normal brain tissues and different grade gliomas based on the gene expression profiles associated with malignant progression of glioma.[Methods ] 1. The DNA repair genes were selected from the gene expression profiles associated with malignant progression of glioma by bioinformatic retrieving GeneBank and GeneCard Database;2. The expression pattern of DNA repair genes were detected in normal brain tissues and different grade gliomas by RT-PCR and Dot-blot.[ Results] 1. 17 up-regulated DNA repair genes were picked out from the gene expression profiles associated with malignant progression of glioma by bioinformatics methods;2. The DNA repair genes , such as Rad17, Rad18 were proved up-regulated in gliomas by RT-PCR; Rad17 was also proved up-regulated obviouslyin gliomas by Dot-blot.[Conclusion] The DNA repair genes , such as Rad17, Rad18 are proved up-regulated in gliomas and associated with the malignant grade of glioma. That may provide us new targets for glioma treatment.
