The genus sargasso of sargassceae comprises 50 species in our country,of which 30 species are commonly seen. As a species of this genus,sargassum fusiforme (Harv.) Setchell is usually called " small-leaf sargassum'',also named Lujiaojian and so on,and distribute the seas in Liaoning,Shandong,Fujian,Zhejiang,Guangdong,et al. Growing wildly and collecting in Spring and Autumn. The whole dried algite is used for drug and called sargassum,wildly used in folk medicine for the treatment of Yingliu,Luoli,swelling and pain of testis. Modern pharmacology studies showed the effects on cardiovascular,increasing the function of immune and macrophage phagocytosis,lowering cholesterol,and also the effects of anticoagulant,ulcer-preventive,antineoplastic,anti-infectious and radiation-resistant et al. The main chemical constituents contain alginic acid,mannitol and iodine et al.As a kind of commonly and frequently encountered disease,diabetes mellitus belongs to endocrine dysfunction diseases,and mainly displayscarbohydrate metabolism obstruction. Up to now,the morbidity have been increasing,WHO has studied and estimated that the patients will grow 122% in whole world in 1995-2025. It's mortality only lower than that of tumor and cardiovascular diseases. So many countries in the world have been paid attention to finding new drugs,especially sieving and studying new hypoglycemic drugs from nature drugs. Many nature drugs and their chemical constituents have been found their hypoglycemic effects. Before our study,there're no reports about the hypoglycemic effect of sargassum fusiforme (Harv.) Setchell,but as folk medicine,it's used to decrease the patient's blood glucose level. In order to explore the pharmaceutical resources,we make research on sargassum fusiforme (Harv.) Setchell,including pharmacognosy identification,pharmacology activity sieving and it's chemical constituents.1. Pharmacognosy identificationBecause the variety and quality have serious influence to our study,we make pharmacognosy identification firstly. As a source of sargassum,the character and diagnostic items are made in Pharmacopoeia. Based on the Pharmacopoeia,we added some physical and chemical identifications,and studied the microfeatures,including the cross section of major axis and thallus,and the power features. The three features are presented after careful examination and comparison.2. Extracting method and Pharmacology activity sievingIn our study,we found the enzyme method was better than the other two methods. We also found the best condition was pH 4.5,10min,12 thousandU/100g,in45Both the alcohol and water soluble fractions of the sargassum fusiforme (Harv.) Setchell processes unnoticeable hypoglycemic effect in normal rats,and also to glucose tolerance in normal or diabetic mice. The alcohol soluble fraction and water soluble fraction showed preventive and hydoglycemic effects to alloxon-induced diabetic mice. In further study,we found the extraction of benzinum,and n-BuOH of alcohol soluble fraction were the main active components which experienced hypoglycemic effects after administration.3. Study on active chemical componentsQualitative test found that the alcohol soluble fraction contains phenols,lactones,and sterid terpenoids,but no alkaloids,flavonoids,anthraquinone and quinoid,and iridoids. Our tests showed benzinum extract contains sterid terpenoids,and n-BuOH extract contains lactones. So the main active components in alcohol soluble faction were sterid terpenoids and lactones.Our study on the water-soluble fraction showed its main component is coarse polysaccharides. The content of the polysaccharides is 56.0% + 2.39%,and its range of molecular weight is from 19.34 X 104 to 21.64X 104. The content of SO42- is 2.47% + 0.25%,Na,Mg and Ca is 1.18% + 0.44%,0.72% + 0.02% and 1.80% + 0.28%. The monosaccharides composed the polysaccharide contains rhamnose,xylose,glucose,fucose,mannose and galactose. The further structural characterization of the polysaccharide is being carried out.
