[Objective] To study the influence of PTEN tumor suppressor gene on expression of VEGF in Malignant Melanoma (MM) cell line;to further understand the mechanism of action of the gene on the malignant melanoma cell line and make a groundwork for exploiting its localization and molecular biological function in the signal transduction pathway of the tumor cells in the future.[Methods] the influence of PTEN tumor suppressor gene on expression of VEGF in Malignant Melanoma (MM) cell line was investigated by using molecular biological,immunological and pathological techniques:(1) We select lipofectin-mediated gene transfection method to create a stable cell line expressing exogenous antisense-PTEN for dual selection. . Using Northern blotting hybridization and Western blotting hybridization method,the expression levels of PTEN tumor suppressor gene were detected in human malignant melanoma cell line after transfection to confirm the expression of target gene;(2) We study theinfluence of PTEN tumor suppressor gene on expression of VEGF in Malignant Melanoma cell line.[Results] (1) Northern blot and Western blot confirmed that the exogenous antisense-PTEN gene expressed in the stable cell line in the presence of ponasterone A;(2) PTEN significantly lowered VEGFexpression in the cells tramsfected with PTEN.[Conclusion] PTEN gene is a clear related suppressor gene of MM. It plays an important role in the mechanisms of the oncogenesis and tumor progression of MM. It is suggested that VEGF participates in the mechanism.
