This thesis is composed of two parts of reseach work .The first one is the expression of ZNF191 gene in malignant ovarian tumour and its clinical significance ,the second is the expression of DNL2A gene in in malignant ovarian tumour and its clinical significance.1. The expression of ZNF191 gene in malignant ovarian tumour and its clinical significance . ZNF191,a novel zinc finger gene of the C2H2 type,was cloned from liver λgt 11 cDNA library by institute of genetics of FU DAN university .This gene is distributed in all kinds of human tissues(apart from heart) , it has been proved this gene has the function of transcription repression ,and may play its role as a negative controler in cell cycles.On these basis,we studied the expression of ZnF191 gene in 31 cases of malignant ovarian tumours and 31 cases of normal ovary tissue by reverse transcription-polymerase chain reaction (RT-PCR) .We found that the expression level of malignant ovarian tumours is much less than that of normal ovary (p<0.01),the expression level of epithelial malignant ovarian tumours is much less than that of granulosa cell tumours (p<0.05), but there is no significant difference among the serousadenocarcinoma , mucinousdenocarcinoma and endometrioid (p>0.05),and that statistical analysis indicated ZnF191 gene expression isn't related to the stage or grade of tumors. These results sugguest the abnormal expression of ZnF191 gene might be involved in the carcinogenesis process of malignant ovarian tumour,and that the loss of ZnF191 gene expression might be a useful predictor of early diagnosis of malignant ovarian tumor,and the results we found may proviod certain new ideal for gene therapy of malignant ovarian tumour .
