Objective To establish subacute senile mouse medol and explore if it can be used as an experimental animal in immunosenesence research,then study the expression of CD137 and it's function on T cells from spleen of aging mouse and subacute senile mouse medol in vitro.Methods Subacute senile mouse model was established by injection of D-galactose. The splenic cells were isolated from mice and cultured in vitro following ConA stimulation. Expression of CD 137 on splenic T cells in the model and normal control groups were compared with each other by flow cytometry method at various cultural times after ConA stimulation. T cells were purified by nylon wool,the supertanant was collected after T cells were cultured for 48h,and stored at -20 C. mRNA in T cells which needed for RT-PCR reaction was isolate by Trizol.Results (1) The CD137 possitve rate on T cells from subacute senile mouse and aging mouse were lower than the normal. The peak of CD 137 expression on T cells from aging mouse appeared earlier compared with normal,and the appearance of the early peak was time dependently with aging.(2) The expression pattern of CD137 from subacute senile mouse was similar to that of the aging mouse.(3) The death ratio of T cells from aging mouse were higher than that of the normal level.(4) mRNA of T cells from aging mouse and subacute senile mouse was decreased,and mRNA from T cells of model mouse seem have a lower quantity than the aging mouse. (5) The female mouse have more tolerantation to D-galactose than the malemouse.Conclusions (1) Subacute senile mouse model can be used in immunosenesence reseach.(2) CD 137 may be an marker of aging.(3) The protection to AICD of CD137 decreased,indicate the function of CD137 was unusual.(4) The effect of D-galactose can modelling the different stage of aging.(5) The decrease of CD 137 expression on T cell from subacute senile mouse model and the aging mouse were due to the declining of their mRNA.(6) The expression of CD 137 on T cells from from subacute senile mouse model and the aging mouse have a time dependently derease and the peak of CD 137 expression appeared earlier while the aging keep going on.
