Objective To investigate the cytotoxicity of peripheral blood mononuclear cells(PBMC) by interleukin-18 combined with interleukin-12(IL-12) and interleukin-2(IL-2),and its mechanism. Methods In vitro incubation of PBMC with IL-18,IL-12 and IL-2(at the concentration of 30ng/ml,10ng/ml,100IU/ml respectively) and Raji cells for five days,detecting the cytotoxicity of PBMC with MTT,measuring the production of IFN-Y,TNF-a .soluble Fas-L(sFas-L) with ELISA,detecting CD16/CD56,perform,granzyme B,Fas-L with flow cytometer. Results IL-18,IL-12JL-2 indepandent or synergistic stimulated the PBMC five days,enhanced the cytotoxicity to Raji cells,and at fourth day,the cytotoxicity was the highest. IL-2 enhanced the production of perforin,granzyme B and proliferation of natural killer cells(NK cells). 11-12 augmented the expression of IFN- Y,TNF- a,perforin,granzyme B,Fas-L from PBMC,enhanced the cytotoxicity of PBMC to Raji cells. IL-18 enhanced the effect of PBMC,augmented the production of IFN- Y,TNF- a,perforin,granzyme B,Fas-L and promoted proliferation of the NK cells. IL-18 increased the proliferation and activation of PBMC induced by IL-18 synergistic with IL-12,IL-2,especially to enhanced the cytotoxicity,augmented the expression of IFN- Y,TNF- a,perforin,granzyme B,Fas-L,they did not rise the sFas-L lever,and the increasing was significant. Conclusion IL-18 enhanced the cytotoxicity of PBMC to tumor cells,augmented the production of IFN- Y . TNF- ?perforin,granzyme B,Fas-L.promoted the proliferation of NK. cells,but didn't increase the expression of sFas-L.IL-18 stimulated PBMC synergistic with IL-12 and IL-2,and increased the cytotoxicity of PBMC against tumor cells appears to be mediated IFN- Y,TNF-u,peribrin,granzyme B,Fas-L.
