Background: Trichophyton rubrum is the commonest cause of dermatophytosis of skin and nail tisssue. A lack of adequate methods for strain identification in T. rubrum has impeded efforts to identify strains endemic to a particular area and determining common sources of infection. However to set up a more simple and practical method for strain identification which is suitable for our country is needed.Objective: 1. Using improved KANE/FISCHER system and develop a standard system for dermatophytes identification. 2. To develop a rapid and reproducible method in strain identification for T. rubrum.Method: 1. The improved KANE/FISCHER system to identify the ATCC strains and clinical isolates of common dermatophytes. 2. Two novel tandem repeat subelements(TRSs), TRS-1 and TRS-2, located in the rDNA nontranscribed spacer(NTS) were amplified from 63 strains of T. rubrum.Results: 1. The improved KANE/FISCHER system can identify the common dermatophytes. 2. Specific amplification of TRS-1 (6 type) and TRS-2(2 type) produced strain-characteristic bandindingpatterns.Conclusions: 1. The improved KANE/FISCHER system can be used as the standard system to identify the common dermatophytes to species level. 2. This new method of typing T. rubrum will provide a rapid, stable molecular method about pathogenesis and epidemiology of this fungus.
