Endometriosis(EMS)is the common ailment within childbearing ages and increasing with the incident rate of 10%. In recent years, EMS has increasing and it can cause dysmenorrhea, chronic pelvic inflammation, pelvic mass, dyspareunia, infertility or acute abdome. The cause and pathogenesis of EMS is still unclear, so treatment is limited and easy to recur. The objective of the study is follow (1)To know the apoptotic of endometrium and peritoneal fluid macrophages in the patients with endometriosis.(2)To evaluate bcl-2,bax and fas genes expression in endometriosis. (3)Add the correlative drugs to the isolated, then observe their apoptotic rate. (4)To analyse the apoptosis and explore the mechanism of Endometriosis and the relative factors further. Methods:(1) We examined eutopic , ectopic endometria and peritoneal fluid macrophages from 22 patients with endometriosis and 14 women with no endometriosis. To obtain evidence for the induction of programmed cell death, apoptotic cells were identified using a modified terminaldeixynucleotidyltransferase-biotin nick end-labelingmethod(TUNEL). To evaluate cell death represser activity, bcl-2,bax and fas genes expression was examined usingimmunohistochemical. (2) We observed their growth and detected their apoptotic rate by FLOW CYTOMETER after the same growth term, the cell of isolated ectopic endometrium and peritoneal fluid macrophages that we cultivated from 12 patients with endometriosis. (3) Observe the change of ectopic endometrium and peritoneal fluid macrophages with the drug under transmission electron microscope.Conclusion:(1) The expression of apoptotic genes were different in eutopic, ectopic endometrium and peritoneal fluid macrophages from EMS and no EMS. Apoptotic rate in EMS was lower than no EMS. Apoptotic rate in EMS was lower than no EMS, and it acceptation was decreased , which could have implications for the growth and survival of ectopic endometrial tissue. (2) Gestrinone and Mifepriston can restrain the ectopic endometrium and peritoneal fluid macrophages and induce them to apoptosis. These medicines can play the important role at the treatment for endometriosis in clinical. (3) the structural change and the morphology of ectopic endometrium and peritoneal fluid macrophages with the drug can be observed under transmission electron microscope.
