[Objective] To compare the depressant effect of the three chemotherapy medicines; arsenic, ADM and DDP and the effects on osteosarcoma cell MG-63 specimens. To approach the depressant effect of combination of arsenic with ADM, DDP on proliferation of osteosarcoma cell MG-63. To research the result when the osteosarcoma cell MG-63 were under the effect of arsenic combined with ADM or (and) DDP.[Methods] The whole experiment was finished in two periods. The methods I used were improved MTT (Methyl thiazolyl tetrazolium) assay. Changes of morphology of arsenic ADM DDP individually or the combination of arsenic with ADM DDP were observed by invert microscope. In the first period experiment, the apoptosis rate of osteosarcoma cell with different concentrations of arsenic ADM DDP individually for 24h, 48h and 72h were tested. Count out the IC50 of the three medicines by SPSS. The second period experiment was on the basis of the data of the first period experiment. The density of ADM and DDP led to 20%-30% osteosarcoma cell to apoptosis and were choosen to be the fixed density when combined with arsenic. Then the apoptosis rate was tested after it had been affected by fixed concentration ADM associated with different concentration arsenic and fixed concentration DDP associated with different concentration arsenic and fixed concentration DDP ADM associated with different concentrations of arsenic for 24h, 48h and 72h. Count out the IC50. The results were analyzed and compared by the data by using SPSS for windows.[Results] Arsenic, ADM and DDP were significantly inhibited by the proliferation of MG-63 cell and the inhibited effect was dose-and-time-dependent. Obvious changes of apoptotic morphology were observed by invert microscope. The result of the first period experiment showed me that after incubation of MG-63 cells with different concentration arsenic ADM and DDP for 24,48,72 hours, the IC50 of arsenic ADM and DDP were 1.92±0.91ug/ml, 4. 76±0. 98ug/ml,5. 89±0. 59ug/ml. The apoptotic rate of arsenic associated with DDP and arsenic associated with ADM, DDP were higher than arsenic associated with ADM(P<0. 05). 1.0ug/ml arsenic, rising up the apoptotic rate of 4.0ug/ml DDP from 18.4% to 37. 5%.[ Conclusion ] Arsenic ADM and DDP all depressed the growth of MG-63 by dose-and-time-dependent. There was no difference among the three medicines when they were used in the same dose and same time. Arsenic could be used to treat osteosarcoma in the clinic as ADM and DDP and could be used associated with DDP. The arsenic associated with DDP could be used as a kind of chemotherapy program.
