IntroductionColorectal cancer is one of the commonest malignant tumors in China and its morbidity is still increasing. Although die operation-based comprehensive therapies have greatly improved the prognosis of colorectal cancer patients in recent years, the postoperative 5-year survival rate, unfortunately, is still about 50%. The two major features of malignant tumors are invasion and metastasis, which are the main death causes of of post-operative patients with cancer. Therefore, researching for valuable biomolecular markers with prognostic significance that are spicific for me invasive and metastatic behaviors could be useful for doctors to predict the invasive and metastatic tendency of colorectal cancer and to give more effective treatments, so that reduces the post-operative recurrence or metastasis and increases the survival rate. At the same time, studies of the molecular biological mechanism of the invasion and metastasis can provide meorectical basis of gene therapy for colorectal cancer.The process of cancer invasion to surrounding tissues and metastasis to distant organs comprises a complex series of sequential steps between tumor cells and host cells interactions, which involve the disruption of homotypic adhesions of tumor cells and the detachment of tumor cells from primary carcinoma, the breakthrough of the basement membrane and extracelluler matrix(ECM) and the penetration into lymphatic and blood circulatioa At die end of this process, circulating tumor cells must extravasation into the host organs. Once malignant cells have arrived at and invaded secondary organsites, they must stimulate local blood vessel growth. The onset of angiogenesis makes neoplastic cells rapid proliferation, local invasion, and ultimately metastasis. Recently, it has been revealed that cell adhesion molecule Syndecan-1 and basic fibroblast growth factor (bFGF) play important roles in the process of the invasion and metastasis.Syndecan-1 is a transmembrane heparan sulfate proteoglycan(HSPG) predominantly expressed on me surface of normal epithelial cells, which can bind several extracellular matrix (ECM) molecules ,other cell adhesion molecules and growth factors via its heparan sulfate chains. Syndecan-1 plays an important role in regulating cell-to-cell and cell-to-extracellular matrix (ECM) adhesion and participating in the modulation of the growth factor response . The extracellular domain of syndecan-1 can inhibit neoplastic invasion and growth. The reduced expression of Syndecan-1 in number of different cancers is closely associated with malignant transformation , aggressive behavior and poor prognosis.Basic fibroblast growth factor (bFGF), a heparin-binding peptide , is a multifunctional cytokine that has pleiotropic roles in many cell types and tissues. As a major angiogenic factor as well as a potent mitogenic factor mainly through a paracrine or autocrine mechanism, it facilitates cell migration and cell proliferation, plays an important role in carcinogenesis and progression, invasion and metastasis. However, to the date time there aren't any reports about combined detection of Syndecan-1 and bFGF expression and their expression related to tumor invasion, metastasis and prognosis in colorectal cancer.In this study, we investigated the expression of Syndecan-1 and bFGF in colorectal cancer tissues by immunohistochemical method(SP) and analyzed the relationship between these markers and clinico-pathological characteristics. We attempted to elucidate the role of the two markers in predicting the invasion, metastatic potential and prognosis of colorectal cancer. Our purpose was to provide useful molecular biological indicators for comprehensive therapies for colorectal cancer.Materials and MethodsIn the present study, 142 patients with colorectal cancer who underwent surgical resection at Zhejiang Provincial People's Hospital from 1991 to 1999 were obtained.
