Two models in vitro and in vivo were established to evaluate the effect of total flavone of pueraria labata (TFP) on diabetic neuropathy (DNP) . The activity of aldose reductase was monitored by determining the content of sorbitol in the red blood cell. Effects of extract of different traditional Chinese medicines on the production of sorbitol were determined . The results showed that TFP, berberine , qercetin , quercetin and Ginkgo biloba extract, isoflavone of soybean could inhibit the production of sorbitol in red blood cells. The effect of TFP was potent. The free radical scavenging activities of extract of different traditional Chinese medicines were studied by determining MDA in homogenate of liver . Results suggested that extact of Radix puerariae, Radix ophiopogonis, Herb epimedii, Radix salviae miltiorrhizae, Fructus crataegi, Radix mori, Rhizoma chuanxiong , Radix, angelicae sinensis, Herba houttuyniae, Ramulus mori, Catharanthus roseus, Herba portulacae and Semen vaccariae could scaveng the free radicals. The effects of extact of Radix pueraria and extact of Radix ophiopogonis were more potent.Diabetic rats were induced by streptozotocin. injected intraperitoneally. After streptozotocin administration, the body weight of diabetic rat was decreased significantly, food and water intake increased. Plasma glucose,glycation ferrohemoglobin increased obviously . The withdrawal latency was prolonged and the latency of licking paw on hot plate was significantly shortened in diabetic rats. The nerve conduction velocity was decreased obviously. Nerve sorbitol content was increased and inositol decreased . Diabetic rats had great abnormal structure in sciatic nerve including paranodal demyelination, axon atrophy , nerve fiber swelling etc.Total flavone of pueraria labata (TFP) administration at dosage of 630 , 1260 , 2520 mg/kg decreased plasma glucose, glycation ferrohemoglobin and nerve sorbitol content and upregulated nerve inositol content obviously. The withdrawal latency of rat was shortened and the latency of licking paw was prolonged in TFP-treated group. TFP could ameliorate morphology abnormalities in sciatic nerve and significantly fasten the nerve conduction velocity. The results suggested that TFP could significantly improve nerve structure and function in streptozotocin rats.Polyol determination by HPLC was improved to determine the content of sorbitol, inositol and glucose simultaneously. The linear equation of sorbitol, inositol and glucose were A = 670176C ( g ) -120463 ( r = 0.9997 ) , A = 907827C ( g ) 70659 ( r = 0.9999 ) , A = 817948C ( Ug ) 305415 ( r = 0.9996), respectively .The CV of them were 2%, 4.1%, 3.4% , respectively. The average recoveries of them were 103.8%, 106.3% , 97.2%. This method is convenient for monitoring the processes of DNP.
