Microcirculation is important place for cell, blood and interstitial fluid to transmit substance, energy and signal. The integrity of function, configuration and metabolism of microcirculation is an important condition to maintain normal function of organs. Microcirculation is sensitive to be injured, Lack of oxygen and nutrition in tissue and cell under microcirculatory disturbance induces function obstacle of organs, which is important base of hypertension, cerebral hemorrhage, hepatitis, tumour, diabetes and many other diseases. More over, continuously existing microcirculatory disturbance aggravates progress and prognosis of diseases. Endothelial cells are main components to form blood vessels of microcirculation, which synthesize and excrete endothelium derived relaxing factor, endothelium derived hyperpolarizing factor and endothelin etc to regulate tensity of blood vessels. As a special incretion organ, endothelial cells have great significance in maintaining cardiovascular homeostasis and normal process of human life. An important pathogeny of microcirculatory disturvances is contracting materials released from endothelial cells to contract microvessels under stimulation of Thromboxane A2 and leukotrienes. Endothelial cell is injured under microcirculatory disturbance and its configuration and function significantly changed. Obviously, there are compact connection between endothelial cells and microcirculatory disturbance. At present, target on endothelial cells to treat microcirculatory disturbance is a perfect strategy.As a basic drug to treat microcirculatory disturbance, anisodamine ( Ani) has been widely used, for example, to treat peptic ulcer, acute high altitude pulmonary edema, migraine, diabetic neuropathy, tuberculous pleurisy etc.One of the key mechanisms of anisodamine to improve microcirculatory disturbance is to vasodilate blood vessels which can antagonize convulsion induced by catecholamines. The mechanism of the vasodilation effect of Ani is not very clear although it was reported to be related to M,a -receptor and calcium-antagonistic effects. The vasodilation effect of anisodamine hasn't been reported to related with endothelial cells. Thus,this experiment aims to study whether the vasodilation effect of Ani relates to endothelium and which material released from endothelial cells to vosadilate blood vessels. To explore whether the vasodilation effect of anisodamine relates with endothelium will supply a new idea to produce more effective drugs to improve microcirculatory disturbance, which has important significance to cure those related diseases. This study was in three parts:I The improvement of anisodamine on microcirculatory disturbance induced by high molecular dextranThe improvement of anisodamine on microcirculatory disturbance induced by high molecular dextran ( HMD ) on mesenteric vascular beds in rats was investigated in this part. The mesenteric vascular bed was placed under the microscope and capillary vessels between 30-45u m were selected and observed. Flying-spots Method was used to measure red blood flowing speed as speed of blood flow, and Autotracking Measurements and Image Camera/Grab was used to measure diameters of blood vessels. After iv injection of HMD (10%) for 1, 3 and 7 min, the microcirculatory disturbance phenomenon was recorded. After iv injection of anisodamine (drug group) or NS (control group) for 1, 3, 5, 10, 20, 30, 60, 90 and 120min, the diameters of vessels, state of blood flow and speed of blood flow were recorded. We found that After iv injection of anisodamine, the speeds of blood flow raised ( P<0.01 ), the blood flow character improved.
