ObjectiveThe study was performed to pretreat with TP and Vit - C, respectively and then injected 25 mol/kg CdCl2 hypodermically. Liver and kidney index , serum activities of GPT and LDH, cadmium level and GSH,MDA concentration, GSH - Px activity in the liver, and renal cortex for the animals were determined 24 hours after the administration of cadmium . We observed the effects of TP and Vit - C on toxicity of cadmium in rats and attempted to explore its possible mechanisms, and bring theory basis for the prevention of cadmium.Methods40 Wistar rats weighting 140 10g, male and female in half were divided into 4 groups at random, 10 in each group. One was control and three groups were experimental. The first group was injected normal saline . The second group was given subcutaneous injection of 25 mol/kg CdCl2. The third groups were pretreated with TP 4mmol/kg, after Ih the rats were given 25 mol/kg CdCl2. and the forth groups were pretreated with Vit - C 500mg/kg, after 2h the rats were given 25 mol/kg CdCl2. After 24hours the rats were weighed, then turned over and abdomen was opened, 5ml blood sample was collected from abdominal aorta, put it quietly for 4 hours, then centrifugal and separate serum. Take serum 0.1ml and GPT in serum was determined by Retimen method. Take another serum 0. 1ml and LDH was determined by 2,4 - Dinitrophenylhydrazine method. After the rats were killed,liver and kidney were taken out after bloodletting, then rinsed and weighed it and made tissue homogenate. Calculate the liver and kidney index respectively. Take 0. 5g renal cortex and liver then threw them into a digestive container make of polytetrafluoroethylene holding 0. 1ml concentrated nitric acid and 2ml of 30% hydrogen peroxide. All tissues were digested wetly for 2 hours at 120 C. Finally the concentration in the digestive fluid was determined by atomic absorption spectrophotometer. GSH concentration was determined by DTNB method, MDA concentration was determined by Thiobarbituric acid method, GSH - Px activity in the liver and renal cortex for the animals were determined by direct DTNB method, Tissue homogenates 0. 5ml were digested with 3M NaOH overnight, the protein was determined immediately after collecting blood samples.ResultsCompared with controls, with cadmium dose of exposure, liver index increased, there was no significant difference in the renal index. Serum GPT and LDH activities increased evidently, Cadmium level in the liver and renal cortex increased, GSH concentration in the liver increased, there was no significant difference in the renal index. MDA concentration in the liver increased. GSH - Px activities in the liver and renal index decreased, MDA concentration has no significant difference in the renal cortex. It indicates that cadmium can increase liver index significantly. It can also increase serum GPT and LDH activities and cadmium level in the liver and renal cortex, It can also increase GSH concentration in the liver and MDA concentration in the liver and renal cortex, especially in the liver. It can check GSH - Px activities in the liver, too. This show liver and renal damage arecaused by cadmium, especially oxidative stress and damage in liver caused by cadmium is serious than those in renal. After pretreated with TP, liver index, LDH activity in serum, cadmium concentration in liver and renal cortex is higher significantly in the rats with cadmium dose of exposure, compared with control. MDA concentration and GSH - Px activity is lower than control. After pretreated with Vit - C, liver index, GPT and LDH activity in serum, cadmium concentration in liver and renal cortex is also higher significantly in the rats with cadmium dose of exposure, compared with control. Serum GPT and LDH activities and GSH, MDA concentration in the rats liver pretreated with TP and Vit - C before cadmium administration were significantly lower than those of the animals given cadmium only.
