PrefaceIR has been the investigative hotspot in medical realm in recently years, as a common risk factor of many pathologic status that result in diabetes hypertension arteriosclerosis lipometabolisic disorder obe-sity. It is resulted from inheritance and environment factors and inter-action between them, Among them the diet is in important position and becomes the main factor that result in diabetes hypertension arterio-scleroisis lipometabolisic disorder obesity etc. In modern times. Feeding rats are similar with changing of dietary composition on devel-opment of insulin resistance in human being, They have become the reasonable animal models that research IR and Euglycemic hyperinsu-linemic clamps techniques are the "Golden standard" that assess IR world wide currently. Now in the research of disease mechanism about cardio - vascular including ( nerve plasma and peptide) have made great progress, especially about NPY CGRP. It has become the hot point in the last decade. In IR the network regulation of nerve and plasma are abnormality, there are vicious circle in diabetes hyperten-sion arteriosclerosis obesity etc. ,So with the research of transforma-tion of NPY CGRP and influence to aorta may break this circulation and found a foundation for therapy.Materials and Methods1. Estabolished in high - fat fed and high - fructose fed in IR in rats-.Sixty adult healthy male Wistar rats (weight 200 -250g)were di-vided into three groups randomly. Each group had twenty rats. Group one was a contrastive group fed with the same energy conventional food, Group two was a high - fructose fed induced group fed with high - rructose diets composition: 69% carbohydrate including ( 34. 5% fructose and 34. 5% cornstarch) 21% protein 10% fat. Group three was a high - fat fed induced group fed with high - fat diets: increasing caloric intake of fat from 10% to 59% , decreasing carbohydrate from 69% to 20%. Four weeks,the whole body decreased more than 50% utilization ratio of glucose and followed with reduced heat - producing fat accumulation, Weight increased clearly. Weight and blood glucose were measured at weekly intervals, After four weeks on diet, weigh blood glucose and insulin levels were measured.2. Euglycemic clamp technique assess IR;By a continuous infu-sion of insulin and glucose , achieved plasma insulin and glucose in balance; In arterial blood, glucose concentration was clamped at the basal fastening level by using a variable glucose infusion rate. The in-fusion was maintained for 120 minutes, glucose and GIR were super-vised constantly per five minute, total 24. Statistics and analysis with three groups and assess IR.3. Examing NPY CGRP and analyzing their meaning in IR rat' s plasma: Examed the rest 45 rats of three groups using radio immune method.4. Computer scanned and quantitated. Software of SPSS 11.5 was used to examined all values.Result1. After four weeks on diets with different composition, show that the weight in experimental groups increased more than that in contrastgroup(P <0.01), In high - fat fed group increase more than that in high - fructose fed group ( P < 0. 05 ). Glucose in rat blood in experi-mental group increased more than that in contrast group (P <0.05).2. Hyperinsulinemic - euglycemic clamp assess that GIR in ex-perimental group is lower than that in contrast group (P <0. 01) ,In high - fat fed group is lower than that in high - fructose fed group ( P < 0.05).3. NPY^ CGRP in rat' s plasma:NPY in experimental group in-creased more than that in contrast group (P <0.01) and it is related to ISI positively, CGRP in experimental group decreased more than that in contrast group (P <0. 01) and it is related to ISI negatively.4. The inner membranes of aorta have been destroyed in IR rats.Conclusion1. After four weeks feeding of high - fat high - fructose dietary composition,the weight of adult male Wistar rats increased obviously, have formed IR.2.
