Blood deficiency which is a common symptom in clinic of Chinese Medicine is due to the inferior spleen and Ventriculus in biochemistry, acute or chronic blood losing also lead to this result. Si-Wu-Tang (Decoction of four ingredients)is famous for enriching and regulating blood. Being a classic traditional Chinese medical formula, Si-Wu-Tang is adept in therapying on blood deficiency. In this thesis, in vitro and in vivo experiment were carried out to systematically observe the compatible mechanisms and the effects of Si-Wu-Tang on enriching blood, and the active substance was studied too.In vivo experiment on the model mice with blood deficiency,targets under below were investigated such as the cells apoptosis of bone marrow and spleen; peripherally hemogram, amount of CD34+ cells; the cell cycle; and also the effects on proliferating colony-forming unit of hematopoietic stem celL progenitor cell in bone marrow were observed.as well. In vitro experiment, by adding with the supernatant of the NIH-3T3 cells induced by Si-Wu-Tang and its different ingredients, the multiplication of NFS 60 cell was observed. Furthermore, exploration was gone deep into pursuiting the active substance of Si-Wu-Tang, To collect all: Si-Wu-Tang is best in all the observing guideline suggests the precise of the formula, And the components take their different effects in various aspects.Such as the supernatant induced by the Radix paeoniae alba correlationgroups could promote the proliferation of NFS60 cell, The best effective group was Radix paeoniae alba with Radix angelicae sinensis. It suggested that Radix paeoniae alba may induce the development of G-CSF or some factors like; Radix rehmanniae with Radix angelicae sinensis or with Rhizoma chuanxiong could promote the multiplication of BFU-E and CPU-Mix without any transparent differences comparing with the full formula, they also have marked effection to CFU-E. So Radix rehmanniae possibly exerts it's action on the multiplication of the erythroid colony and CPU-Mix; The Radix angelicae sinensis correlatin groups could increase the amount of white blood cell in peripherally hemogram, and promote the proliferation of hematopoietic stem cell and progenitor cell; Radix paeoniae alba with Rhizoma chuanxiong group hase minus effect on rising the amout of BFU-E colony; The groups which have Rhizoma chuanxiong exert their action on depressing the apoptosis of marrow bone cells and spleen cells, they could regulate the cell cycle as well.In analysising the effective foundation of Si-Wu-Tang, the pursuit researchment is on comparising and selecting the most active constituents ,while substances and active components are segregating at the same time.The result shows that the active substance of Si-Wu-Tang may be include paeoniflorin, some ploysaccharides, monosaccharides,disaccharides or some other ingredients with the similar polarity.Through cluster analysising the result of the full formula and it's all groups,the below conclusion could be given: Radix paeoniae alba is the important drug to promote the proliferation of NFS-60 cell; and has conteraction on BFU-E; Radix angelicae sinensis could increase the amount of white blood cell, also action on the proliferation of different hematopoietic progenitor cell ; Radix rehmanniae takes it's major function on the multiplication of CFU-E and CPU-Mix.The research of Si-Wu-Tang formula is on macrocosm, organs, cells and molecule from different levels. All the studies on Si-Wu-Tang are aim to primarily clarify the compatible mechanism and it's functionary base thus to establish a foundation for the modernization of Chinese tranditonal medicines.
