Objectives: To investigate the concentration of plasma plasminogen activator inhibitor-1 and the promoter - 844 G→A substitution, -675 4G/5G polymorphism in patients with unstable angina pectoris and control.Methods: 121 patients with unstable angina pectoris (UAP group) and 108 patients with angiography-proved no stenosis(control group) were studied. Plasma PAI-1 antigen was measured by ELISA. The segment of PAI-1 gene was amplified by PCR(-675 4G/5G and -844 G→A). The products of PCR were digested by Xho I ( - 844 G→A). Results: No difference in genotype frequency was observed in both groups. Plasma level of PAI-1 antigen in UAP group was higher than that in control group (68.15±45.29 ng/ml vs.53.39±20.62 ng/ml, P<0.001) and related to the 4G/5G and G→A polymorphism. Carriers of A or 4G allele have high plasma PAI-1 antigen level. Plasma PAI-1 antigen level was significantly associated with the number of diseased vessels, Glu , ApoB, TC and LDLc.Conclusion: PAI-1 plays an important role in regulation of the fibrinolysis function. In patients with UAP the PAI-1 antigen was much higher. Promoter -844 G→A substitution and -675 4G/5G polymorphism have no relationship with the occurrence of UAP.
