| Isatin (2,3-dioxoindoline) is an endogenous inhibitor of monoamine oxidasetypeB (MAO-B) . Monoamine oxidase is the main metabolism enzyme of monoamines , and MAOs have important physiological functions in the body. Parkinson's disease (PD) is a kind of neural disease closely related to MAOs. Many evidences have shown that the activity of MAO-B in PD patients is upregulated significantly. We investigated the inhibition effect of isatin in rats, the urine content of isatin in health people and PD patients determi-ned by high performance liquid chromatography(HPLC), and a clinical test in 16 PD patients. The results are as follows:1 The rats were feeded with isatin for 6 months, the blood samples of the rats was determined, the content of MAO-B in tested rats reduced significantly. (P<0.01)2 The urine content of health people and PD patients indicated that: (1) The quantity of isatin in urine is increased with age (P<0.05) . (2) Urinary isatin levels in PD patients significantly increased compared with healthy group. (PO.01) (3) There is no significant difference among the PD patients in different stage.(P>0.05)3 According to the method of Webster, the progressive rate was compared between the tested and control groups. From the result we can see that the PD symptoms of 5 patients among 7 improved slightly in tested group, and 5 patients among 9 aggravated slightly in control group. Because there were only 16 patients were tested , there was no significant difference in statistics. But isatin can improve the symptom of PD patients, which implies that isatin may has the prophylaxis and treatment effects inPD.The results suggested that isatin, the endogenous inhibitor of MAO-B, could significantly inhibit the activity of MAO-B in tested rats, and could be studied as the marker of PD for early-warning, prophylaxis and treatment.
