Isatin also named 2,3-indole is known as a new kind of specific inhibitor of MAO-B in brain and it also exists in sea organisms. MAO-B is the major enzyme in the metabolism of monoamine neurotransmitters and it accounts for eighty percent. The metabolism rate of DA is intensely related to the inhibitor of MAO-B. The increased metabolism of DA will make more output of free radical which can make the dopaminergic neurons dying to develop the Parkinson's Disease(PD). The study on the MAO-B inhibiting mechanism of 2,3-Indole is done to learn the mechanism of PD and find a new drug to treat PD.Objective: The experiment is done to study the effect of 2,3-Indole on the content and release of the monoamine transmitters in brain. The effect on the serpateon modek melanocyte and kindling model of 2,3-indole is also done. All these is done to learn more about the Pharmacological activity and anti-PD mechanism of 2,3-Indole.Methods: The Wistar mice are given injection of 50-200mg.kg-1(ip) and killed two hours later through microwaves. Then the corpus striatum are separated in the Glowinski's way to determinate the Ach and DA.The Wistar mice are injected 2,3-indole 200mg.kg-1(ip) and following give mice serpaton lmg.kg-1(sc) to observe the trembling reaction. Using the UVB and 2,3-indole on the single cell suspension of preduce in normal adult, then respectively to detect the activity of tyrosinase and content of melanin. Dissolve the 2,3-indole in solution of DMSO, then give 25,50,100,160,200mg.kg-1 to the kindled rats and the given volume is 1ml.kg-1. After Discharge Threshold(ADT) is examined 30 minutes later and record the Discharge Latency, After Discharge Duration and seizure grade.Results:1 The concentration of Ach and DA in striatum both are increased after giving rats 2,3-indole 50 or 200μmol.l-1(ip). The changes of DA have a dose-dependent relation with drug dosage. 2,3-indole can promote the release of DA, 5-HT and NE especially the effect on DA is significant(p<0.05,p<0.01).2 2,3-indole 200mg.kg-1 has no effect on the formation procedure of serpaton model.
