Objective To detect the levels of interleukin-2 and interleukin-6 in the serum, and in intraabdominal fluid of patients with endometriosis. To study on the expression of interleukin-2, interleukin-6 in ectopic endometrial tissue.Methods The levels of interleukin-2 and interleukin-6 in the serum, and in the intraabdominal fluid of 30 cases with endometriosis were detected using enzyme linked inmmunoabsorbent assay( ELISA) and compare with the counterparts of 20 cases without endometriosis(control) . The ectopic endometrial tissue from the same patients with endometriosis and the endometrial tissues of 10 normal women have been investigated for the expression of interleukin-2 and interleukin-6 by inmunohistochemistry method.Results (1)There was no statistically significant difference between the level of interleukin-2 in the serum and in the intraabdominal fluid between patients with endometriosis and the control group (P>0.05). The level of interleukin-2 hi the serum and in the intraabdominal fluid of patients with endometriosis, statistical analysis showed no significant difference between early stages and advanced stages.(2)Significantly elevated levels of interleukin-6 (cytokines of T help cell 2) were found in the serum and intraabdominal of patients with endometriosis(P<0.05). There was statistically significant difference between the level of interleukin-6 detected in the serum and hi the intraabdominal fluid of those in early stages( I > II stage) and those in advanced stages(IIL IV stage).The levels of interleukin-6 of serum and intraabdominal fluid in patients with advanced stage were higher than those in early stage (P<0.05).(3)The ratio of interleukin-2/ interleukin-6 decreased in the serum and in the intraabdominal fluid between patients with endometriosis and the control group(P<0.05). (4)The levels ofinterleukin-6 detected in the intraabdominal fluid of patients with endometriosis have a positive correlation with those of in the serum (rs=0.745 , P<0.01) . The levels of interleukin-6 in the patients with endometriosis have a negative correlation with the IL-2/IL-6 in the serum and in the intraabdominal fluid respectively.(rs=-0.406, rs=-0.48 P<0.05). (5)Interleukin-2 and interleukin-6 were expressed in the interstitial cells of ectopic endometrial tissue. The expression ratio was56.67% ,60% respectively .There was statistically significant difference in the expression of interleukin-2, interleukin-6 between the ectopic endometrial tissue and endometrial tissue. (6) The levels of interleukin-2, interleukin-6 in the patients with endometriosis and control group have no significant correlation with menstrual cycle.Conclusion (1)The levels of interleukin-6 in the serum and in the intraabdominal fluid of patients with endometriosis are increased. Interleukin-6 plays an important role in the pathophysiology of endometriosis. The levels of interleukin-6 of serum and intraabdominal fluid hi patients with advanced stage were higher than those in early stage. (2) The ratio of mterleukin-2/interleukin-6 decreased in the serum and hi the intraabdominal fluid between patients with endometriosis and the control group .It imply Thl cell shift to Th2 cell in patients with endometriosis. (3) The levels of interleukin-6 detected in the intraabdominal fluid of patients with endometriosis have a positive correlation with those of in the serum. (4)Stronger expression of interleukin-2 and interleukin-6 in the ectopic endometrial tissues may contribute to the disturbed immune regulation in patients with endometriosis.Postgraduate Li Jianxia(Gynecology)Directed by Prof Dai Shuzhen...
