Objective To evaluate the expression and distribution of tumor necrosi factor-a (TNF-o)in the lung tissue of chronic obstructive pulmonary disease(COPD) model of rats and the relationship between TNF-o and the airway inflammatory reaction. The effect of Ambroxol on increasing the output of pulmonary surfactant and alleviating airway inflammatory reaction were also investigated.Methods Wistar rats were randomly divided into three groups. The normal group, The model group and the ambroxol treated group. The COPD model of rats was established by intratracheal instillation of lipopolysaccharide twice and daily exposure to cigarette smoking. The group was treated with ambroxol since the first day through. Pathologic changes of the lung tissue and blood gas analysis were performed. MLI,MAN were measured by computer image analyzer. Also the expression of TNF-ot were determined by in situ hybridization.Results Compared with the normal group, the model group presented significant differences in PCO2,PO2 ,MLI and MAN [(53±4)mmHg vs(41±3. 5)mmHg,(80. 5± 8. 6)mmHg vs(98. 4±2. 9)mmHg,(92±10)/玭 vs(52±3)ptm,(268±15)/mm2 vs(410± 27)/mm2]P<0. 01; The expression of TNF-a were also markedly elevated[(0. 318 ± 0. 031)vs(0. 197±0. 036)]P<0. 01. In comparison with the model group, blood gas analysis in the ambroxol treated group were improved [PCO2 (48±3mmHg vs(53±4)mmHg, PO2(89. 2±7. 3)mmHg vs(80. 5±8. 6)mmHg] P<0. 01. The expression of TNF-a were decreased [ (0. 282±0. 043)vs(0. 318±0. 031)]P<0. 05 and MLI.MAN were significantly different [(83±8)/xm vs(92±10)/im,(291±10)/mm2 vs(268±15)/mm2] P<0. 05.Concluson Ambroxol could show its protective effect in COPD model of rats by alleviating airway inflammatory reaction and increasing the output of pulmonary surfactant.
