Panax Ginseng is a well-known Chinese traditional medicine in "invigorating qi". Saponins of Panax Ginseng (SPG) is one of the main effective fractions extracted from Panax Ginseng. Our previous study had suggested that total saponins of Panax Ginseng (TSPG) can markedly promote the proliferation and differentiation of murine CFU-S,BFU-E,CFU-E,CFU-GM and CFU-MK in vivo and ex vivo。In this study, by using the techniques of culture of hematopoietic progenitor cell in semisolid agar ex vive, biological assay of activities of hematopoietic growth factors in the culture media of bone marrow stromal cells, endothelial cells, monocytes, thymocytes,and splenocytes induced by TSPG, the effect of TSPG on the proliferation and differentiation of CFU-GM in normal human and relative mechanism are studied. The purpose is to study the effect of TSPG on the proliferation and differentiation of human granulocyte-macrophage progenitor cells, and to clarify the modern biological mechanism and provide the experimental evidence for clinic application of Panax Ginseng. Our results indicated that: 1,after culturing bone marrow mononuclear cells in semisolid agar for 10 days, the grown cell balls are identified through the techniques of Wright' staining, histocytochemistry and immunocytochemistry as CFU-GM. 2,with the adding of granulocyte-macrophage colony stimulating factor (GM-CSF), TSPG can markedly promote the proliferation and differentiation of CFU-GM. 3,the conditioned culture media of bone marrow stromal cells, endothelial cells, monocytes, thymocytes and splenocytes induced by TSPG can also enhance the colony formation of CFU-GM at different level. Take all results together, we speculate that TSPG may directly and/or indirectly stimulate bone marrow stromal cells such as endothelial cells, monocytes and fibroblasts in hematopoietic inductive microenvironment(HIM) and lymphocytes and induce them to produce or secrete GM-CSF and other hematopoietic regulating factors which in turn promote the colony formation of CFU-GM. This may be one of the modern biological mechanisms of Panax Ginseng promoting hematopoiesis.
