Objective To study the anti-tumor effect of Swelling-Dispersing Decoction in vivo and in vitro,and to clarify the mechanisms of the anti-tumor effect of the decoction,and to indicate the anti-tumor effect of sijunzitong.Methods Li vivo experiment:Establish mice bearing neoplasm (SRS-82 cell line) as the experimental tumor model,then use Chinese traditional medicine decoction feed the mice,the 5-FU group was given by 5-Fluorouracil abdominal injection. After the treatment of fifteen days,kill the mice. Stripping and weighing the neoplasm block,calculating the inhibition rate of the anti-tumor. We use electronic microscope to observe the neoplasm cells,and extract the DNA of the cells for agarose gel electrophoresis. In vitro experiment:culture the SRS-82 cell line,choose the fourth or sixth generation as the experimental object. Use the already prepared blood serum contain medicine interfere in the cells,then we detect the cell vitality by MTT Technique,assay the apoptosis rate of the cells using the flow cytometer. We also adopt immunocytochemistry technique to sense the expression intensity of bcl-2 protein,and in situ hybridization cytochemistry technique to detect the MMP-2 mRNA expression intensity.Results l.The weights of the neoplasm of medicine-feed groups are significant lighter than the control groups'(P<0.01). The neoplasm block's weight of 5-FU group are the lightest(P<0.01). 2.Under the electronic microscope,we find that the tumor cells of Swelling-Dispersing Decoction group appear special alteration which infer apoptosis:nucleus karyopyknosis,Fragmentation,the structures and envelope of organelle are Integra on the whole. Still in this group,the agarose gel electrophoresis of the DNA educe ladder figure. 3. Both Swelling-Dispersing Decoction group and 5-FU group's cell vitality is significant lower than that of control group(P<0.01). The 5-FU group's cell vitality is the lowest. 4. By the FCM,we detect that the apoptosis rate of Swelling-Dispersing Decoction group is significanthigher than that of control group. 5. The expression intensity of bcl-2 protein of Swelling-Dispersing Decoction group is obviously lower than that of control group(P<0.01). 6. The expression intensity of MMP-2 mRNA of every group have no distinct difference.Conclusion Swelling-Dispersing Decoction and SiJunZitong both has obvious anti-tumor effect in vivo. The former can also inhibit tumor cell proliferation in vitro. The Swelling-Dispersing Decoction is likely to induce the tumor cells apoptosis. The mechanism of this decoction inducing apoptosis may be that it could reduce the expression of the bcl-2 gene,which is considered to repress apoptosis. Both the Swelling-Dispersing Decoction and the SiJunZito has no influence on the expression of MMp-2 mRNA of the tumor cells.
